Embodiment Construction

[0046] The present invention will be further described below in conjunction with the examples, but not limited thereto.

[0047] 1. Transcriptome data sources

[0048] The transcriptome data of American squash come from the results of Illumina high-throughput deep sequencing of the fruit by our research group in 2016. The samples were entrusted to Beijing Biomike Biotechnology Co., Ltd. for RNA-Seq transcriptome sequencing, and 83,650 Unigenes were assembled through the De Novo method as analysis data.

[0049] 2. Transcriptome SSR site identification and SSR primer design

[0050]Use the software MISA to search for SSR sites in the loofah transcriptome, the search criteria are: repeat unit length 2~6 bp, dinucleotide, trinucleotide, tetranucleotide, pentanucleotide and hexanucleotide The minimum number of repetitions was 6, 5, 5, 4, and 4, respectively. The Primer 3.0 software was used to design and evaluate primers for the sequence before and after the SSR repeat unit, an...

Abstract

The invention discloses an SSR primer group developed on the basis of a zucchini transcriptome sequence and application of the SSR primer group in identification of a genetic relationship and a hereditary character of zucchini, and belongs to the technical field of molecular biology. The SSR primer group comprises 35 pairs of primers, and nucleotide sequences of the first pair of primers to the 35th pair of primers are as shown in SEQ ID No. 1 to SEQ ID No. 70. The obtained SSR primer group has the advantages of abundant polymorphism, good repeatability, stable amplification, clear electrophoretic bands and the like. Meanwhile, because the primers are from zucchini expression genes and can reflect variation of a functional domain in a genome, diversity of genetic germplasm of loofah can be distinguished well. The primer group can be used for zucchini material resource genetic diversity analysis, genetic relationship identification, DNA finger-print construction and the like.

Description

technical field [0001] The invention specifically relates to an SSR primer set developed based on the sequence of the American pumpkin transcriptome and its application, and belongs to the field of biotechnology. Background technique [0002] Pumpkin ( Cucubita pepo L. ) is an annual vine herb of Cucurbitaceae Cucurbitaceae, and one of the earliest cultivated crops by humans. It is delicious, nutritious, and has medicinal value and is loved by the public. In the production of American squash, it has strong adaptability. The planting cycle is short, the cultivation is relatively easy, the product yield is high, the supply cycle is very long, and it is also relatively easy to store and is suitable for intercropping. It has a wide range of planting in my country and has become one of the main varieties of melon vegetables in my country. First, the cultivated area is second to cucumber. The development time of American squash breeding in my country is relatively short, and t...
