Abstract

The invention relates to a tumor metastasis inhibitor of a targeted mouse platelet membrane glycoprotein GPIb alpha. The tumor metastasis inhibitor is a monoclonal antibody 1D12 or 2B4. The inventionfurther relates to an identification method of the tumor metastasis inhibitor of the targeted mouse platelet membrane glycoprotein GPIb alpha. The targeting performance and functionality of the inhibitor are verified by establishing verification models of the tumor metastasis inhibitor of the targeted mouse platelet membrane glycoprotein GPIb alpha. Through verification of the four verification models, it is determined that the targeted mouse platelet GPIb alpha monoclonal antibody 1D12 or 2B4 can effectively inhibit tumor cell metastasis and does not influence normal platelet functions. The possibility is provided for the research of the specific action mechanism of GPIb alpha in the tumor metastasis process.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to pharmacology, and specifically refers to a tumor metastasis inhibitor targeting mouse platelet membrane glycoprotein GPIbα and an identification method thereof. Background technique [0002] Platelets can promote tumor metastasis, and the first and key point in promoting tumor metastasis is the activation of platelets by tumor cells to form tumor thrombi. Various adhesion molecules on the surface of platelets, such as glycoprotein (Glycoprotein, GP) Ib-IX-V, GPIIb-IIIa, etc., interact to mediate the adhesion of tumor cells to platelets. Among them, GPIbα is the largest and most important component of the GPIb-IX-V complex. Studies have shown that knocking out the GPIbα gene or replacing the GPIbα extracellular region in mouse models can significantly reduce the lung metastasis of lung cancer in mice, but it will be complicated by thrombocytopenia (Jain S, Zuka M, Liu J, et al.Proc N...
