Abstract

The invention discloses a wheat grain color related CAPS marker and a detection method thereof and relates to the technical field of molecular breeding of wheat. The CAPS marker is named as KU4104-6AL, and has a sequence of SEQ ID NO. 1. A method for identifying the wheat grain color comprises the following steps: 1, extracting DNA of to-be-detected wheat genome; 2, performing PCR (Polymerase Chain Reaction) amplification on the DNA of the wheat genome by utilizing sequences such as SEQ ID NO. 2 and SEQ ID NO.3 so as to obtain an amplification product; 3, performing digestion on the amplification product by utilizing a NruI incision enzyme having a recognition sequence of TCG / CGA so as to obtain enzyme-digested products; 4, ensuring that the color of corresponding wheat grains is dark whenthe enzyme-digested products are 214bp and 212bp, and naming genotype of the corresponding amplification product as KU4104-6A-a; ensuring that the color of corresponding wheat grains is light when the enzyme-digested product is 426bp. The CAPS marker KU4104-6AL developed by the method in the invention is capable of effectively distinguishing colors of the wheat grains and is used for genetic improvement of sprouting resistance of wheatears and flour quality.
