Embodiment 1

[0051] To investigate the effect of fluridone added at different growth stages of Schizochytrium on the content of oil and fatty acids in Schizochytrium. When the Schizochytrium was cultured to 0h, 24h and 48h, 50mg / L fluridone was added respectively. After 120 hours of fermentation, the bacteria were collected and analyzed for each component. Such as figure 1 As shown, the addition of fluridone has no significant effect on the biomass of Schizochytrium; in terms of oil content, adding fluridone at 0h can significantly reduce the accumulation of oil, and adding fluridone at 24h is beneficial to the accumulation of oil , adding fluridone at 48h had no obvious effect on the oil accumulation of Schizochytrium; in terms of EPA content, the effect of adding fluridone at different times on the fatty acid composition of Schizochytrium was shown in Table 1. The addition of fluridone All can increase the content of EPA, and the effect of adding it at 24h is the best, and the content ...

Embodiment 2

[0055] In order to investigate the effect of fluridone on the whole growth stage of Schizochytrium, 50mg / L fluridone was added after the culture of Schizochytrium until the 24th hour, samples were taken every 24 hours, and the biomass, oil and EPA content were analyzed .

[0056] Depend on figure 2 and image 3 It can be seen that after the addition of fluridone, the growth of Schizochytrium and the synthesis of oil are delayed, and the oil and biomass of the control group are decreased at 48h, 72h, and 96h, but there is no significant difference with the control group at 120h , and at 144h, compared with the control group, the oil was significantly improved. Such as Figure 4 It was shown that the addition of fluridone was beneficial to the accumulation of EPA in the whole process, and at 144 hours, the proportion of EPA in fatty acids reached 0.75%, an increase of 42.31% compared with the control group. Adding fluridone at the early stage of Schizochytrium's oil synthes...

Abstract

The invention provides a regulation and control method for improving the EPA (Eicosapentaenoic Acid) content in schizochytrium and application and relates to marine microorganisms. A strain schizochytrium sp. MYA1381 is subjected to flat plate and second-grade seed activation to prepare a seed solution; then the strain is inoculated to a fermentation culture medium; fluridone is added at differentphases of the growth of the schizochytrium. The fluridone can be applied to improvement of the EPA content in the schizochytrium. The fluridone is applied to regulation and control of a fatty acid synthesis way for the first time and a new thought is provided for improving the EPA content in the schizochytrium. The fluridone is added and the content of EPA in schizochytrium grease is remarkably improved and is improved by 42.31 percent when being compared with a manner without the fluridone. A foundation is provided for improving the EPA content in the schizochytrium by utilizing the fluridone.

Description

technical field [0001] The invention relates to marine microorganisms, in particular to a control method and application for increasing EPA content in Schizochytrium. Background technique [0002] Polyunsaturated fatty acids (PUFA) refer to fatty acids with two or more unsaturated double bonds and more than 20 carbon atoms in their molecular structure. According to the distance between the double bond and the methyl end, it can be divided into ω-3 series, ω-6 series and ω-9 series polyunsaturated fatty acids. Among them, ω-3 series polyunsaturated fatty acids have physiological functions such as preventing and treating cardiovascular and cerebrovascular diseases, promoting the normal growth and development of brain tissue and retina, and are essential fatty acids for human health (Gill and Rao 1997; Das. et al. 2003). The omega-3 series polyunsaturated fatty acids mainly include docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA). The molecule of EPA is the formula ...
