[0050] clinical sample

[0051] In the screening phase, the whole blood of 6 breast cancer patients and 6 age-matched healthy controls collected at Qilu Hospital of Shandong University from February 21, 2012 to July 9, 2012, were detected by miRNA microarray. In the validation phase, from June 27, 2016 to August 30, 2017, Qilu Hospital of Shandong University recruited 68 breast cancer patients and 13 healthy controls. All patients and controls were Han females.

[0052]Fasting venous whole blood (2 ml) was collected and untreated whole blood samples were immediately stored at -80°C or in liquid nitrogen until further analysis. All breast cancer blood samples were collected before surgery or any treatment. Control samples were collected from healthy women with no past or current history of malignancy or inflammation. All participants obtained written informed consent. This study was approved by the Ethics Committee of Qilu Hospital of Shandong University.

Abstract

The present disclosure relates to a miRNA marker for diagnosing breast cancer and the application of miRNA thereof, the markers are miR-26b-5p, miR-106b-5p, miR-142-3p, miR-142-5p, miR One or a combination of ‑185‑5p and miR‑362‑5p; miR‑26b‑5p, miR‑106b‑5p, miR‑142‑3p, miR‑142‑5p, miR‑185‑5p, and miR The sequence information of ‑362‑5p is shown in SEQ ID NO.1~6, respectively, and it is worth noting that the combination of miR‑185‑5p and miR‑362‑5p shows a higher AUC than any single miRNA in the 6 miRNAs , sensitivity and specificity.

Description

technical field [0001] The disclosure belongs to the field of biomedicine, and in particular relates to a miRNA marker for diagnosing breast cancer and the application of the miRNA. Background technique [0002] The statements herein merely provide background information related to the present disclosure and may not necessarily constitute prior art. [0003] Breast cancer is the most common tumor in the world and the leading cause of cancer death in women. At present, there are many studies on the early detection of breast cancer using various biomarkers such as tissue protein, nucleic acid and plasma protein, but their real clinical application is very limited. Therefore, it is very important and urgent to develop novel non-invasive biomarkers for detection of breast cancer with high sensitivity and specificity. [0004] MicroRNAs (miRNAs) are small non-coding RNAs that regulate gene expression at the post-transcriptional level by degrading or inhibiting the translation o...
