Abstract

In optical-based sample analysis, eg fluorescence-based or absorbance-based measurements, a choice is made between a first excitation light path and a second excitation light path. A first excitation optical path directs excitation light from the light source through the excitation monochromator, through the excitation filter and to the sample. A second excitation light path directs excitation light from the light source through the excitation filter and onto the sample while bypassing the excitation monochromator. The excitation light generated by the light source is transmitted along the first excitation light path or the second excitation light path according to the selection made, thereby illuminating the sample. In response, the sample produces emitted light (transmitted light in the case of absorbance measurements), which is transmitted to and measured by a photodetector.
