[0173] Certain tumors are particularly difficult to control with conventional therapies. Hypoxia is a characteristic feature of solid tumors in which cancer cells exist at very low oxygen concentrations. The hypoxic area often surrounds necrotic tissue and as a solid form of cancer grows too large for its vasculature. When the vascular supply fails to meet the metabolic demands of the tumor, the tumor microenvironment becomes hypoxic. Multiple regions within tumors contain <1% oxygen compared to 3-15% in normal tissue (Vaupel and Hockel, 1995), and avascular regions can constitute 25-75% of tumor mass (Dang et al., 2001 ). Approximately 95% of tumors are hypoxic to some extent (Huang et al., 2004). Systemically delivered anticancer agents rely on tumor vasculature for delivery; however, poor vascularization hinders oxygen supply to rapidly dividing cells, making them useful for targeting cell proliferation in poorly vascularized hypoxic tumor regions.
