Problems solved by technology

However, the identification of common cattle and zebu by this method is based on the deletion of small-segment genome sequences. Compared with the deletion identification of large-segment genome sequences, the conditions are more stringent, false positives are more likel

Abstract

The invention discloses a genomic sequence capable of being used to distinguish z Bos Indicus and Bos Taurus bloodlines. The genomic sequence is as shown in SEQ ID NO. 1 and 2, and can be used to distinguish Bos Indicus and Bos Taurus and facilitate the implementation of later cattle breed pure breeding, improvement and breed conservation works, Bos Indicus lacks the SEQ ID NO. 1 genomic sequence, and Bos Taurus is normal. The invention also discloses a molecular biology method for distinguishing Bos Indicus and Bos Taurus. The method comprises the following steps: 1) separating and extractingDNA from a tissue of the cattle to be identified; 2) using a forward primer (SEQ ID NO .3) and a reverse primer (SEQ ID NO. 4) to perform PCR amplification; 3) taking the amplified product and performing electrophoresis in a 1.5% agarose gel; and 4) observing the electrophoresis result, if a band is amplified as SEQ ID NO. 2, it can be judged that the cattle to be identified is Bos Taurus, otherwise the cattle to be identified is Bos Indicus. The genomic sequence of the invention can be used as an effective target for identifying bloodlines of existing cattle breeds, and is used to identify Bos Indicus and Bos Taurus, a necessary resource and method are provided for bloodline origin analysis and hybrid improvement of cattle breeds, and the identification method is simple and convenient, accurate and effective.

Description

technical field [0001] The invention relates to the fields of species identification and molecular biology, in particular to a genome sequence that can be used to identify zebu and common bovine lineages and an application method thereof. Background technique [0002] Common cattle (Bos Taurus) and Zebu (Bos Indicus) are the two main subspecies of cattle, accounting for more than 90% of the world's existing cattle stocks, and are the main sources of important livestock products such as milk and beef in the world. Play a leading role in the world cattle industry. There are obvious differences between common cattle and zebu in terms of geographical origin and breeding distribution. Common cattle mostly originated in Europe, while zebu cattle originated in India and other Asian countries. There are also research reports that Africa is also the origin of common cattle and zebu cattle. In terms of geographical distribution, ordinary cattle mostly live in the northern regions wh...
