Abstract

The invention discloses development and application of agave cpSSR marker primers. The cpSSR marker sequences corresponding to SEQ ID NO.21 to SEQ ID NO.40 of the primer pair are developed based on sequencing of a chloroplast genome of an agave cultivar H.11648, the developed cpSSR marker sequences and a homologous sequence of a published chloroplast genome species of agave are subjected to multiple comparison analysis, polymorphic cpSSR site screening and cpSSR marker primer design are carried out, and polymorphic primers are applied to agave resource evaluation through sequencing and comparison of PCR products. According to the patent, the problem of low development efficiency of a traditional cpSSR polymorphism marker is solved, and the blank of agave cpSSR marker information at presentis filled; and the developed cpSSR polymorphic marker primers lay a foundation for research of systematic classification, genetic diversity evaluation, germplasm identification and the like of the agave.
