Problems solved by technology

[0003] However, the integrity of the skin of patients with severe burns is poor. As an invasive skin test, there are many difficulties in the clinical application of blood tests for such patients. Repeated blood testing will also aggravate the suffering of patients.

Embodiment 1

[0021] Example 1 Collection and processing of urine specimens

[0022] The burn patients were selected as the burn group, and the healthy subjects during the same period were selected as the normal control group. 30ml fresh morning urine samples were collected from the subjects in each group after admission. Those who could not urinate normally were collected from their morning catheters and placed in a dry and clean container. After the collected urine samples were centrifuged at 4000 r / min for 5 min, the supernatant was aspirated, and each tube was divided into 2 ml and stored in a -80 °C refrigerator.

Embodiment 2

[0023] Example 2 Mass Spectrometry and Screening of Urine Peptides

[0024] Urine samples were extracted for protein and the concentration of the extracted protein was determined. Mass spectrometry analysis of urine samples was performed by OrbitrapFusion mass spectrometry. The data obtained in the mass spectrometry of the experimental group and the normal control group were quantitatively calculated. Differences between groups were analyzed by t-test, and the differentially expressed proteins were screened with the difference in protein expression more than 1.5 times and the statistical test P<0.05 as the reference standard.

Embodiment 3

[0025] Example 3 Identification and Analysis of Differential Peptides

[0026] The database used was Uniprot_Homo database, and the original mass spectrum files were processed by MaxQuant software. See Table 1 for search parameter settings.

[0027]

[0028] Compared with healthy people, talin 1 is highly expressed in the urine of burn patients, such as figure 1 As shown, the main biological processes it participates in are as follows figure 2 As shown, the expression of talin 1 in the urine of the normal control group and the burn group was significantly different.

Abstract

The invention provides application of a urine ankle protein 1 (Talin-1) and a polypeptide fragment thereof, and particularly relates to application of the urine ankle protein 1 and the polypeptide fragment thereof in preparing preparations for burn diagnosis, differential diagnosis, burn area and degree evaluation, treatment effect evaluation, monitoring, prognosis evaluation, mechanism research and the like. Burns are common important wounds in daily life, about 5,000-100,000 people in every one million people are burnt every year, according to statistics of World Health Organization, more than 300,000 patients die from burns globally every year, and the treatment survival rate of seriously burnt patients is still at a lower level. Researches prove that compared with healthy people (a normal control group), the expression of the urine ankle protein 1 and the polypeptide fragment thereof in burn patients is increased, and the urine ankle protein 1 and the polypeptide fragment thereof can be used for various purposes of application detection of burn patients. The advantages of noninvasive acquisition, large-scale repeated sampling and convenient preservation of a urine sample are exerted, and the urine sample is used for detecting the urine ankle protein 1 and the polypeptide fragment thereof.

Description

technical field [0001] The invention relates to new uses of urine talin 1 and its polypeptide fragments, in particular to urine talin 1 and its polypeptide fragments in burn diagnosis, differential diagnosis, burn area and degree evaluation, treatment effect evaluation, monitoring, prognosis evaluation and mechanism applications for research, etc. Background technique [0002] Burn refers to the damage of human skin, tissue and even deep organs caused by chemical substances such as flame, high temperature gas, hot solid or liquid, radiation, electricity, strong acid and alkali. It is a systemic comprehensive disease. Mass necrosis, infection, shock and coagulation dysfunction of wound tissue after burn can cause a series of pathophysiological changes in the body. Different degrees of burns have different effects on the human body. Severe burns can cause damage to the internal environment of the human body, causing burn patients to experience complex pathophysiological chang...
