Problems solved by technology

This technology is simple and has a large amount of coding, but each microsphere needs to correspond to an optical fiber, and the cost of thousands of tiny spheres requires hundreds of microfibers, which cannot be directly detected by the existing detection instruments of conventional lab-on-a-chip , adding an additional cost

Embodiment Construction

[0021] The fabrication and detection of the binary coded microsphere self-assembled array biochip of the present invention are made by four steps: (1) surface modification of microsphere carrier and in situ synthesis of coding sequence and probe sequence; (2) microsphere carrier Self-assembly on the membrane plate and self-assembled ordered microsphere arrays are transferred and fixed to the glass plate. (3) Decoding: relying on two different states of the physical properties of the microspheres and the presence or absence of hybridization signals between decoding molecules and coding sequences A set of binary numbers identifying the probes on each microsphere. (4) The target molecule is hybridized with the probe on the microsphere, and the information of the target molecule is determined according to the hybridization signal.

[0022] The specific preparation process is as follows:

[0023] Microsphere carrier 2 can be pearlescent dye, fluorescent microsphere, magnetic micro...

Abstract

The invention relates to an array type biology chip based on microballons carrier and its coding-decoding method. The said chip comprises a microball carrier, poly dimethyl siloxanes layer, glass sheet, code molecular, nucleic acid and protein probe molecular. The microball carriers are ararranged in order, and the coding molecular and the probe molecular are coupled onto the microball carriers in covalent. The coding decoding method is to use the different states of the pearly dye as the microball carrier, either fluorescence microball or the magnetic ball with identical physical property but different two states which metches with the presence or not of the hybrid signal of the coding series to form a binary number to code the particles. Finally, whether there is target molecular existence or not in the sample can be determined by the hybrid signal with the probe series.

Description

technical field [0001] The invention is a simple and convenient high-throughput detection gene chip for nucleic acid detection of biological samples and a coding and decoding method for the chip. Background technique [0002] Nucleic acid is the genetic material in organisms, the carrier of genetic information, and has species specificity. Physiological and pathological phenomena in organisms are directly or indirectly related to genes. Therefore, gene detection is widely used in scientific research and clinical diagnosis, as well as identification of numerous biological species and individuals, and is a basic tool in the above-mentioned fields. Since the functions of various genes are closely related, the development of high-throughput gene detection technology has far-reaching significance. Among the existing high-throughput gene detection technologies, there are mainly position-encoded array biochips with glass slides as carriers and liquid-phase biochips with fluorescen...
