Embodiment 1

[0036] 12 g of an epoxy-containing methacrylate polymer with an average particle size of 35 microns in diameter and 250 ml of dioxane were placed in a 1 L round bottom flask equipped with a funnel, stirrer, reflux condenser and positive nitrogen pressure inlet, Stir for 30 minutes to swell the resin. 20 g of polyethyleneimine (PEI, average molecular weight 600 Daltons) was added and the funnel was rinsed with a further 150 ml of dioxane. The stirred mixture was refluxed overnight. After the mixture had cooled, the mixture was transferred to a suction filter flask, drained, washed once with dioxane, three times with methanol, and dried under vacuum at 60°C. Elemental analysis: 57.8% C, 7.4% H and 5.6% N.

Embodiment 2

[0038] 5 g of methacrylate polymer containing epoxy groups and 150 ml of dioxane were placed in a 500 ml round bottom flask equipped with a stirrer and reflux condenser. 30 g of PEI (average molecular weight 1200 Daltons) was added and the mixture was refluxed overnight. After cooling, the mixture was transferred to a suction filter flask, sucked dry, washed twice with dioxane and three times with methanol, and dried. Elemental analysis: 55.7% C, 7.8% H, 5.5% N.

Embodiment 3

[0040] 5 g of epoxy-containing methacrylate polymer and 150 ml of a 50 / 50 mixture of dioxane and water were placed in a 500 ml round bottom flask equipped with a stirrer and reflux condenser. 30 g of PEI (average molecular weight 1200 Daltons) was added and the mixture was refluxed overnight (89°C). After cooling, the mixture was transferred to a suction filter flask, sucked dry, washed twice with dioxane and three times with methanol, and dried. Elemental analysis indicated that the final product contained: 57.5% C, 7.4% H, 2.5% N.

Abstract

This invention concerns the preparation and use of novel polymeric chromatographic media and preferably mixed mode polymeric chromatographic media. In accordance with the present invention, polymeric media is prepared using polymeric particles derivatized with polyethyleneimine, and preferably such polyethyleneimine derivatized polymeric particles further functionalized with appropriate reactants. The polymeric chromatographic media is especially useful for bioseparations.

Description

field of invention [0001] The present invention relates to the preparation and use of novel chromatographic media, preferably mixed polymeric chromatographic media, for the separation and purification of various biomolecules such as peptides, proteins and antibodies. More specifically, the present invention discloses novel chromatographic media, preferably mixed anion exchangers, mixed anion-cation exchangers and hydrophobic exchangers. The chromatographic media are prepared by modifying the polymer with polyethyleneimine and functionalizing it. It was unexpectedly found that these mixed polymer media provided enhanced separation and protein binding capabilities. Background of the invention [0002] The analysis of protein mixtures by ion-exchange chromatography media is well documented in the literature, eg, Pete Gagnon, "Purification Tools for Monoclonal Antibodies", Validated Biosystems, Inc., (1996). More recently, the development of protein-based pharmaceuticals and v...
