AI Technical Summary

Benefits of technology

This patented technology helps isolate specific types of bacterial sporas that cause damage or harm crops like Chinese cabbage (Brassica oleracea). These germs are found naturally throughout indoor environments but they may also be introduced into other areas where crop production needs to protect them against insect attacks.

Problems solved by technology

This patented describes how certain types of bacteria can be controlled by natural agents like moldings or yeast that produce neurotoxic substances called Bacillanidium spp., which helps fight these harmful organisms. However, there may also exist some specific species of blue-green algae (Bacillaria dendrodes), another type of microorganism commonly known as sea urchin shellfishes, causing damage when they infest crops such as rice plants during their growth period. These naturally occurring biocontrol mechanisms cannot prevent this problem because they lack selective pressure against all three important classes of nematodes: phytophagous ones, germinating weeds, and feed on them.

Embodiment 1

[0011] 1.1 Source of materials

[0012] The carcass of a Bemisiatabaci infested by fungus fungus was collected from vegetable cucumbers in Guangzhou area.

[0013] Potato dextrose agar medium (PDA): boil 200g of potatoes to extract the juice, then add 20g of glucose + 17-20g of agar powder to the extracted juice, add water to make the total volume 1000ml and boil. Afterwards, it was sterilized by autoclaving (121°C, 30min).

[0014] Aseptic operating conditions: All utensils and utensils must be sterilized by autoclaving (121°C, 30min), and operations such as inoculation are carried out in ultra-clean workbenches.

[0015] Culture conditions: Culture in a 25°C light (14L: 10D) incubator. After colonies are formed, transfer to a PDA slope, and then transfer to a 4°C refrigerator for storage.

Embodiment 2

[0057] Example 2 Determination of the pathogenicity of Paecilomyces fumigatus PF01-N10 strain to Bemisia tabaci

[0058] Bioassay is one of the effective means to detect the degree of lethality and lethality rate of entomogenic fungi to target pests, and can provide an important reference for comprehensive evaluation of the biological control potential of the fungus. The invention measures the pathogenicity of the Paecilomyces fumosorose PF01-N10 strain to the whitefly, so as to screen out the optimal concentration used in the control.

[0059] Insects and host plants to be tested: Bemisia tabaci, adults of Bemisia tabaci raised on cucumbers in the greenhouse, inoculated to insect-free cucumber seedlings, after 12 hours of oviposition, remove adults, and place cucumber seedlings at 25±0.5°C In the light incubator of the whitefly, it will be used when the Bemisia tabaci develops to the 2nd instar nymph. Cucumis sativus L., a variety of Wanji cucumber, was purchased from the Ve...

Embodiment 3

[0070] The pathogenicity determination of embodiment 3 Paecilomyces fumigatus PF01-N10 strain to diamondback moth larvae

[0071] Insects and host plants for testing: Plutella xylostella, adults of Plutella xylostella raised on cabbage in the net room, inoculated on the seedlings of cabbage without insects, after 12 hours of laying eggs, remove the adults, and place the seedlings of cabbage at 25 ± In a light incubator at 0.5°C, wait until the diamondback moth develops to the 2nd instar larvae. Chinese cabbage Brassica campestris L.ssp.chinensis was purchased from Vegetable Research Institute of Guangdong Academy of Agricultural Sciences, and potted seedlings with 6-8 expanded leaves were used for the test seedlings.

[0072] (1) Treatment of test strains

[0073] The Paecilomyces fumigatus PF01-N10 strain obtained after purification was cultivated on a PDA plate in a thermostat (14L:10D) at 25±0.5°C for 10 days, and collected by adding 0.3% Tween-80 sterile water For spores...

Abstract

The invention discloses a paecilomyces fumosoroseus strain. The strain is a wild strain separated from bemisia tabaci body in Guangzhou area, which is naturally infected by worm living fungus. The wild strain is re-planted on the bemisia tabaci body for the rejuvenation, so that the paecilomyces fumosoroseus strain is obtained; the purified strain is obtained through single spore isolation. The paecilomyces fumosoroseus strain is PF01-N10; the preservation number of the strain is CCTCC No: M207088; the strain is stored in China Center for Type Culture Collection on 27 June, 2007. Through a long term of infectious biology research and the indoor biology testing, the paecilomyces fumosoroseus has strong infectious and killing effects for a plurality of sucking mouth parts insects, such as the white fly and the cotton worm etc, and the chewing mouth parts.
