Problems solved by technology

[0005] At present, there is no report in China about the research on grassland fescue anther culture. Although there are reports in the world, only albino seedlings have been obtained (Rose et al., 1987). Albino seedlings cannot survive and cannot be used in production

Embodiment 1

[0037] Embodiment 1, preparation and statistical observation of haploid plants of "Grassland Fescue No. 17"

[0038] 1. The acquisition of anther-induced callus and its statistical observation

[0039] 1. Anther acquisition and low temperature pretreatment

[0040] Get the uninucleate mid-stage anthers of the "Grassland Fescue No. 17" (seeds purchased from Keluowo Grass Industry Co., Ltd.) plant normally grown in the greenhouse.

[0041] Insert the collected grass fescue tassels (with anthers inside) into a triangular flask filled with tap water, cover them with plastic bags, bring them back to the laboratory and put them in the refrigerator, and store them under low temperature and dark conditions for 7 days at 4°C. Then microscopic examination corrects the appropriate age of anthers: peel off the young spike inflorescences of the meadow fescue inflorescences after the low-temperature pretreatment, and peel off the anthers from the upper, middle and lower parts of the inflor...

Embodiment 2

[0081] Embodiment 2, preparation and statistical observation of No. 1 haploid plant of grassland fescue

[0082] The difference between this embodiment and Example 1 is that the explant-anther that adopts is the anther of the uninucleate late stage of Fescue No. 1 (fescue seeds are purchased from Keluowo Grass Industry Co., Ltd.) plant, and all the other steps are the same as those in Example 1. 1's agreement.

[0083] The experimental statistical observation result of the present embodiment is as follows:

[0084] 1. Statistical observation of callus induced by anthers

[0085] The experimental results are shown in Table 6 below, and the average callus induction rate can reach 77.3%.

[0086] Table 6. Induction rate of callus

[0087]

[0088] 2. Differentiation rate of regenerative differentiation culture

[0089] The experimental results are shown in Table 7 below. It can be seen from Table 7 that the average differentiation rate of callus can reach 34%, and the aver...

Abstract

The invention discloses a meadow fescue anther tissue culture method and a special culture medium thereof. A provided differentiation culture medium of the meadow fescue is a solid culture medium prepared by adding the following substances to MS (Murashige And Skoog) basic culture fluid: kinetin, NAA (Naphthalene Acetic Acid), a carbon source and a gelling agent, wherein the final concentration of the kinetin is 1.5-2.5 mg / L, and the final concentration of the NAA is 0.4-0.6 mg / L. The method of the invention comprises the following steps of: putting the meadow fescue anther on a callus tissueinduction culture medium to be cultured so as to obtain induced callus tissue; and then, transferring the callus tissue to the differentiation culture medium of the invention for differentiation culture to obtain haploid regenerated plants. The callus tissue induced by the meadow fescue anther can be effectively differentiated by the differentiation culture medium to obtain regenerated plants. Byutilizing an androgenesis technology in the method, regenerated plants with a homozygous genetic background is obtained, and the selection efficiency is improved.

Description

technical field [0001] The invention relates to the fields of plant tissue culture and crop breeding, in particular to a method for grassland fescue anther tissue culture and a special culture medium thereof. Background technique [0002] Grassland fescue, scientific name Festuca pratensis L, also known as meadow fescue, is distributed in Eurasia and North America in temperate regions, and is also distributed in Northwest and Southwest my country. It is a perennial herb. Dense fibrous roots, erect stalks, smooth, plant height 70-130cm. The leaves are flattened into involutes, 15-20cm long, the auricles are lanceolate, sickle-shaped and curved, with eyelashes on the edge. Panicle tight or slightly stretched, 10-25cm long, spikelets 10-15cm long, containing 6-10 flowers, glumes lanceolate to ovate, 3-5mm long, glabrous, margin membranous, lemma long ovate , 5-8mm long, apex glabrous, ovary glabrous, anthers up to 4mm. It can adapt to a wide range of soils, and grows best in...
