Problems solved by technology

However, there is a major bottleneck in the successful use of this broadly applicable new technique: how to selectively capture and enrich target exomes in a head-to-tail step from scattered genome, mitochondrial and other forms of DNA in an efficient manner. intron or target intron region

Abstract

In one aspect, the present invention relates to a method for selectively capturing and / or amplifying exons or targeted genomic regions from biological samples. In one embodiment, the method includes the steps of obtaining DNA templates for the targeted genomic region, cloning the DNA templates into cloning vectors to form template DNA clones, constructing libraries of the template DNA clones at least comprising the targeted genomic regions, generating hybridization probes from the DNA template clones in the libraries, capturing the targeted genomic DNA regions by hybridizing the targeted genomic DNA samples (fragmented either mechanically or enzymatically) with the generated hybridization probes, and eluting the captured genomic fragments by using conditions for releasing and separating the bound DNA from the hybridization probes.

Description

technical field [0001] The present invention generally relates to the capture and amplification of targeted genomic DNA (gDNA) regions, and more particularly, the present invention relates to the selective capture and amplification of DNA and / or RNA probes from any biological species ( Methods that include all exons, any subset of exons, or any other region of interest of the genomes of animals, plants, fungi, protozoa, archaea, and eubacteria), mitochondrial and other forms of DNA. Background technique [0002] Current technological developments have enabled deoxyribonucleic acid (DNA) sequencing at the scale of millions to billions of base pairs (bps) per experiment (Margulies et al., 2005). At least three companies now offer commercial massively parallel sequencing systems (for example, the 454 system from Roche; the Illumina system from Illumina; the SOLiD system from Applied BioSystems). At present, the sequencing capabilities of some of these systems are sufficient fo...
