[0111] 1. In the following experiments, the antiproliferative effects of different tocopherols and tocotrienol isomers were compared using melanoma cell lines. All 8 vitamin E isomers were found to inhibit the proliferation of malignant skin cancer cells. Potency varies between the various isomers, with γ-T3 being the most potent. A detailed study of the effect of γ-T3 revealed that γ-T3 treatment of two melanoma cell lines resulted in the induction of apoptosis, which was associated with the downregulation of pro-survival factors such as Id-1 or EGFR. Meanwhile, activation of JNK and inactivation of NK-κB were also observed after treatment. Inhibition of JNK activity by a specific inhibitor (SP600125) partially blocked sensitivity to γ-T3 treatment, suggesting that γ-T3-induced apoptosis is mediated through the JNK signaling pathway.
