Abstract
The invention relates to an Ames test method for biologically evaluating cigarette tipping paper, belonging to the technical field of tobacco. The method of the invention comprises the following steps of: taking artificial saliva as tipping paper leaching liquor; quantitatively taking the tipping paper with a certain area, and shearing into a strip form in 1cm*6cm, and placing in a 50-250mL conical bottle with a grinding stopper; injecting the leaching liquor in the conical bottle according to a proportion of a water filling area / the leaching liquor being 24-36 cm<2> / mL, covering the stopper; placing the conical bottle in a bath oscillator with temperature of 37+ / -1 DEG C for leaching under oscillation for 5-180min to prepare sample raw solution; doping the sample raw solution in a plane plate with dosage of 10muL, 50muL, 250muL, 500muL and 1000muL, respectively counting reverse mutation bacteria colony counts of TA97, TA98, TA100 and TA102; and finally evaluating the safety of the cigarette tipping paper according to reverse mutation test results of salmonella typhimurium. The method provided by the invention can effectively simulate actual conditions of the tipping paper contacting with the oral cavity to prepare the sample; the sample raw solution for the Ames is suitable for dosage range; and the method is practical and simple and convenient.
