Abstract

The invention relates to a method for separating and identifying tuckahoe strains by using dry tuckahoe. The method comprises the following steps: (1) separating strains by using tuckahoe sclerotium, namely pretreating dry sclerotium, promoting growth of hypha, and taking a block for inoculation to obtain tuckahoe hypha; and (2) culturing and identifying the hypha, namely selecting a culture medium, inoculating, culturing, directly observing under a microscope, ensuring that the tuckahoe hypha grows in a symmetrical dichotomous branching form on the top under the natural condition, thus obtaining the tuckahoe strains. By the method, that whether the separated strains are high in quality can be judged according to the texture of the dry tuckahoe, and a blank that the later quality cannot be accurately predicted when the strains are separated through fresh tuckahoe is filled up. In addition, the method is feasible, easy to implement, accurate in identification and simple, and special instruments are not required. The dry tuckahoe is taken as a separating material, and the tuckahoe strains can be successfully separated; and moreover, the tuckahoe strains can be accurately identified according to the discovered natural growth phenomenon of tuckahoe.
