Embodiment 1

[0020] An experiment to detect the activity of the compound methyl thiobutyrate on Caenorhabditis elegans by two-grid method:

[0021] 1. Cultivation and preparation of nematodes

[0022] Caenorhabditis elegans was inoculated on oat medium (20 g of oat, 80 mL of water), and cultured at 28°C for 5-7 days. The required nematodes were washed out with the Baermann funnel method, and the nematodes were diluted to 1000 / mL with sterile distilled water as a nematode suspension.

[0023] 2. Test method

[0024] Add water agar to each of the two grids, add 1 mL of nematodes (about 1000) to the surface of the water agar in one grid, add 10 μL and 40 μL of the pure compound to the other grid, and seal with Parafilm. Place them at room temperature, and check and calculate nematode mortality under an optical microscope at 6, 12, and 24 hours respectively.

[0025] The method of identifying death: dead nematodes are stiff or in a "J" shape, and live nematodes are curled and twisted. Immo...

Embodiment 2

[0033] This example uses the same experimental agent and the same experimental method as Example 1, except that the nematode tested is replaced by Meloidogyne incognita. The experimental method for detecting the monomeric resistance of the volatile compound methyl thiobutyrate against Meloidogyne incognita by the two-grid method is as follows:

[0034] 1. Cultivation and preparation of nematodes

[0035] Tomato roots were used for the preparation of the southern root-knot nematode (Meloidogyne incognita). Take the root system of tomato plants with severe disease, wash the root system, count the number of root knots, continue soaking in water overnight, and collect the soaking liquid; soak and wash 3 times in the same way, and discard the root system. The residual liquid and soil were centrifuged at 2000r / min for 2min, the supernatant was discarded, 494g / L sucrose was added, and after mixing, the supernatant was cleaned with an 800-mesh separation sieve, and the larva and larv...

Abstract

The invention relates to volatile compound methyl thiobutyrate and application thereof, and belongs to the technical field of a microbial biological pesticide. The volatile compound methyl thiobutyrate is detected by the following steps that a, the used bacterial strain is achromobacter (Achoromobacter sp.) CD-253, and the preservation number is CGMCC No6358; b, cultivating a CD-253 bacterial strain in an LB culture medium by using a conventional fermentation method, and carrying out headspace extraction on a fermentation liquor by using a solid-phase microextraction technology; c, carrying out gas chromatography-mass spectrometer (GC-MS) analysis on an extracting matter; and d, carrying out comparison of a national Institute of standards and Technology (NIST) database to obtain volatile metabolite methyl thiobutyrate. The volatile compound methyl thiobutyrate has inhibiting effects on caenorhabditis elegans (Caenorhabditis elegans) and meloidogyne incognita (Meloidogyne incognita), and can be applied to preparation of a vermifuge preparation and a precusor substance. Compared with the traditional nematicides pesticide, the volatile compound methyl thiobutyrate has the advantages of no environment pollution, low cost and excellent nematode killing effect.

Description

Technical field: [0001] The invention relates to a volatile compound methyl thiobutyrate and an application thereof, and belongs to the technical field of microbial biopesticides. Background technique: [0002] Plant nematode disease is a serious plant disease, and it is estimated that the global annual crop loss caused by plant parasitic nematodes reaches 157 billion US dollars (Abad et al.2008). At present, chemical control is still one of the important measures to control plant nematodes, but the long-term use of highly toxic chemical insecticides is increasingly harmful to the environment and human health. Therefore, bionematicides have become a research direction for nematode control. Microorganisms are easy to cultivate and have a short growth cycle, so the use of microorganisms and their metabolites to control nematodes has attracted people's attention. [0003] The bacterial strain used in the present invention is Achromobacter sp. CD-253, and the volatile substanc...
