Abstract

The invention discloses a method for in vitro culturing of a regenerated plant of dendrobium officinale. The method comprises the following steps: (1) pretreating materials, namely, darkly culturing a dendrobium officinale tissue culture seedling for 48 hours to obtain a tender stem tip; (2) inducing a callus and stripping off the stem tip, and inoculating the stem tip on a callus induction culture medium to be cultured; (3) inducing an adventitious bud and taking the well-grown dendrobium officinale callus, namely, transferring the dendrobium officinale callus to an adventitious bud induction culture medium to be cultured to obtain a clustered adventitious bud; and (4) carrying out subculture, namely inoculating the clustered adventitious bud to a subculture medium to carry out the subculture, wherein culture conditions are as follows: culture is carried out at a constant temperature of 25 DEG C for 30 hours, the illumination culture is carried out for 16 hours each day, and the illumination intensity is 2000lx and the dark culture is carried out for 8 hours. The technical scheme disclosed by the invention is an efficient and safe breeding way. The adventitious bud is induced by a tissue culture technique and the intermediate propagation problem of the dendrobium officinale can be solved.
