Embodiment 1

[0024] Embodiment 1: the evaluation of benzydamine hydrochloride anti-influenza virus activity

[0025] 1. Experimental materials

[0026] 1.1 Cells, viruses and drugs

[0027]MDCK cells were purchased from American Type Culture Collection (ATCC); virus strain: A / PuertoRico / 8 / 34 (H1N1); drug: benzydamine hydrochloride (CAS: 132-69-4) was purchased from Sigma.

[0028] 1.2 Experimental Instruments

[0029] Multi-labeled microplate reader (PerkinElmer).

[0030] 2. Experimental methods and results

[0031] 2.1 Cell culture: 37°C, 5% CO 2 cultured in a humidified incubator. A DMEM medium containing 10% FBS, 100 U / mL of penicillin and streptomycin was used. Cells were subcultured to 90% confluence, and the subculture ratio was 1 / 3–1 / 4.

[0032] 2.2 Virus culture: Take SPF chicken embryos aged 9 to 11 days, check them with an egg tester before inoculating the virus, and mark them at a position far away from the embryos, disinfect and punch holes and inoculate 2 4 The virus ...

Embodiment 2

[0042] Example 2: Evaluation of the broad-spectrum anti-influenza virus effect of benzydamine hydrochloride

[0043] On the basis of Example 1, this example further tested the antiviral activity of benzydamine hydrochloride on influenza A virus subtype H6N6 strain, influenza A virus subtype H7N8 strain and influenza B virus.

[0044] 1. The virus strains used in the experiment include: A / Duck / Hubei / 5 / 2010(H6N6), A / Duck / Hubei / 216 / 1983(H7N8), B / human / Huber / 1 / 2007

[0045] 2. Divide MDCK cells into 1.5×10 4 Cells / well were seeded in 96-well cell culture plates, cultured in a 37°C cell culture incubator for 14-18 hours, and the cells were grown into a single layer before use. Discard the medium in the well plate, wash twice with PBS, add 100TCID 50 Virus fluid (100 μl) and different concentrations of drugs (100 μl) were cultured in a cell culture incubator at 37°C. The initial concentration of the drug was 50.0 μM, and 6 gradients were serially diluted 2 times, and two replicat...

Abstract

The invention relates to the technical field of medicines and in particular discloses an application of benzydamine hydrochloride in preparing a medicament for preventing or treating influenza virus infections. Firstly, the toxicity of benzydamine hydrochloride for an MDCK cell is detected, and a result shows that the maximum nontoxicity concentration of benzydamine hydrochloride is 50.0 mu M; secondly, the antiviral activity of benzydamine hydrochloride is measured in the safe and nontoxic concentration range, and a result shows that the small molecule compound has remarkable antiviral activity and the antiviral effect has a dose-dependent effect; finally, the antiviral activity of benzydamine hydrochloride for different types and subtypes of influenza viruses is detected, and a result shows that benzydamine hydrochloride can inhibit replication of all detected influenza virus strains in a dose-dependent manner and the anti-influenza virus activity of benzydamine hydrochloride has broad spectrum. Therefore, the invention discloses benzydamine hydrochloride as a novel broad-spectrum anti-influenza virus medicament for the first time. Benzydamine hydrochloride can be used for preparing the medicament for preventing or treating influenza virus infections.

Description

technical field [0001] The invention relates to the technical field of medicines, in particular to the application of benzydamine hydrochloride in the preparation of medicines for treating or preventing influenza virus infection. Background technique [0002] Influenza viruses belong to the family Orthomyxoviridae, the genus Influenzavirus. According to the differences in the antigenic and genetic characteristics of the virion nucleoprotein (NP) and matrix protein (M), influenza viruses are divided into three types: A, B, and C, also known as A, B, and C. The whole genome of influenza A virus consists of eight single-stranded negative-strand RNAs of different sizes, named after segment 1 to segment 8, respectively. The full-length viral genome is about 13.6kb, encoding 10 structural proteins (PB2, PB1, PA, HA, NP, NA, M1, M2, PB1-F2 and NS2 / NEP) and non-structural proteins (NS1). Influenza A viruses can be further divided into 17 H (H1-H17) and 10 N (N1-N10) subtypes based...
