[0063] 1.1.1 Preparation of medium

[0064] Add glutamine to Ham's F12 (GIBCO, Carlsbad, Calif.) to a final concentration of 2mM; penicillin-streptomycin to a final concentration of 100U / ml and 100μg / ml; fetal bovine serum (FBS, GIBCO, Carlsbad, Calif.) To a final concentration of 10% (volume / volume), culture medium A containing FBS and penicillin-streptomycin was obtained, stored at 4°C, and freshly prepared once every two weeks.

Abstract

The invention discloses a CHMM1 culture medium suitable for in-vitro culture of Chinese epidermal melanocytes. The CHMM1 culture medium takes a base culture medium suitable for in-vitro culture of the epidermal melanocytes as a matrix, wherein the matrix is added with the following components: (1) serum with the volume concentration of 5%-20%, (2) 1-100 ng/mL of bFGF and 1-100 ng/mL of GM-CSF, and (3) 1-100 [mu]g/mL of 3-isobutyl-1-methylxanthine (IBMX) and 3-3000 ng/mL of alpha-MSH. The invention provides the culture medium suitable for in-vitro culture of the Chinese epidermal melanocytes, and the culture medium can be used for culture of the epidermal melanocytes having strengthened proliferation and melanin synthesis abilities, so that the epidermal melanocytes are better applied in transplantation treatment of skin disease patients with hypopigmentation or no pigments.

Description

(1) Technical field [0001] The invention relates to a CHMM1 medium suitable for in vitro culture of Chinese epidermal melanocytes. (2) Background technology [0002] The change of human skin color is directly related to the number, size, type and distribution of melanosomes. Skin color is primarily determined by the combination of pigments manufactured by the skin and natural pigments contained in the top layers of the skin. The epidermis on the surface of the epithelial layer of the skin contains epidermal melanocytes that produce skin pigment (melanin). In people with hypopigmented or amelanotic skin conditions, the melanocytes in the skin are missing, damaged, or nonfunctional, resulting in white spots on various parts of the body. [0003] Melanin is a protein derivative produced by melanocytes in the epidermis. At present, it is believed that epidermal keratinocytes are involved in the formation and metabolism of skin melanin. Melanocytes and keratinocytes are collec...
