Problems solved by technology

During the counting process, most workers will dilute the samples. However, the samples must be counted within the effective time, otherwise, air bubbles will appear in the counting frame due to water evaporation, causing measurement errors and affecting the accuracy of counting.

Embodiment 1

[0025] The present invention will be described in detail below in conjunction with the accompanying drawings.

[0026] The invention provides a novel sealing method for quantitative sample identification of phytoplankton and protozoa, and its complete structure is as follows: figure 1 As shown in the figure, a 0.1ml ZH7116 type plankton counting frame 1, a cover glass 2, a paper strip 3 with a length of 26 mm and a width of 2 mm, and a glass slide 4. The length and width of the paper strips are determined according to the size of the counting frame, so as to coincide with the cover glass 2 and achieve the best sealing effect.

[0027] A kind of sealing method of phytoplankton, protozoan quantitative sample identification, its steps are:

[0028] 1. Use scissors to cut 80g of A4 paper into paper strips with a length of 26mm and a width of 2mm, dip it in glycerin (1.5ml), place it on the glass slide 4, and repeat 3 times, so that under the action of gravity, the glycerin will ...

Abstract

The invention discloses a mounting method used in a quantitative sample survey of phytoplankton and protozoon. The mounting method comprises the first step of taking a piece of paper and conducting dip-dyeing on the paper with glycerinum, placing the paper on a glass slide and repeating the operation of dip-dyeing for 2-3 times, smearing the glycerinum uniformly and fully on a cover glass, and using residual glycerinum to conduct dip-dyeing on scraps of the paper; the second step of diluting quantified phytoplankton and protozoon samples in body of water after the samples are concentrated, taking diluent and placing the diluent within a counting cell with a volume of 0.1 ml, shaking the glass slide so that the samples can be tiled within the counting cell; the third step of taking the cover glass and conducting covering on the samples in a direction of 45 degrees, and gradually ejecting air in the counting cell; the fourth step of using the scraps of paper which is already dipped and dyed by the glycerinum in the first step to conduct sealing around the cover glass; the fifth step of conducting counting on the samples at a room temperature. If no bubble appears in the samples of the counting cell, then a mounting effect with an optimum efficiency is reached.. The mounting method used in the quantitative sample survey of the phytoplankton and the protozoon is simple to operate, low in cost, and can be reutilized; meanwhile, objective lens of a microscope is not polluted, and effect of the mounting is remarkable, thus working efficiency is improved and precision of sample counting is improved.

Description

technical field [0001] The invention belongs to the field of water body ecological research, and more specifically relates to a sealing method for quantitative sample identification of phytoplankton and protozoa, which is suitable for identification of quantitative samples of phytoplankton and protozoa in water bodies such as reservoirs, lakes, rivers, and ponds. Advanced pre-processing can save the time of repeating the film and improve the accuracy of the technique. Background technique [0002] Phytoplankton are photoautotrophic organisms that live on planktonic life in water bodies and are producers of water bodies. Their individual lifespan is usually very short, from a few hours to tens of days, and they can reproduce many generations in one growth period. Due to their small size, phytoplankton respond quickly to environmental changes and are one of the important indicators for monitoring water eutrophication. As the primary consumers of water, protozoa are a class o...
