Embodiment 1

[0021] Example 1: Isolation, screening and identification of Bacillus LM3403

[0022] Bacterial colonies with obvious morphological differences were isolated and purified from the wild yak manure samples grown on the alpine non-polluted grassland above 3000 meters above sea level by dilution coating method, numbered and stored for future use.

[0023] The plate confrontation method was used to screen the bacterial strains with broad-spectrum antagonistic ability against mold from the bacterial strains obtained from the above separation and purification, and the strain with the strongest broad-spectrum antagonistic ability was obtained, which was named LM3403.

Embodiment 2

[0024] Embodiment 2: Bacillus LM3403 species identification

[0025] Refer to the "Common Bacteria System Identification Manual" for strain morphological observation and physiological and biochemical characteristics analysis. The colony of strain LM3403 on the NA plate is round, opaque and milky white, sticky, and the edges are irregular; the cells are rod-shaped, Gram-positive, spore-shaped, and oval. The colony of strain LM3403 on the NA plate is round, opaque and milky white, sticky, and the edges are irregular; the cells are rod-shaped, Gram-positive, spore-shaped, and oval. Aerobic growth, exercise, positive V-P test, positive catalase test, positive indole test, positive starch hydrolysis, positive casein hydrolysis, positive gelatin liquefaction, positive glucose fermentation test, positive nitrate reduction, positive urea utilization. The morphological characteristics, physiological and biochemical characteristics of the strain LM3403 were consistent with the standard...

Embodiment 3

[0028] Embodiment 3: Bacterial strain LM3403 antibacterial spectrum

[0029] Using Fusarium graminearum, Fusarium moniliforme, Aspergillus flavus, Aspergillus niger, Aspergillus gray-green, Aspergillus fumigatus, Penicillium oxalicum, Penicillium extensa, Mucor tallum and other main harmful molds for grain storage as indicator bacteria, the confrontation culture method was used to determine the strains The broad-spectrum antibacterial ability of LM3403, the results are shown in Table 2.

Abstract

The invention belongs to the technical field of grain storage, and discloses a bacillus subtilis strain having broad-spectrum antagonism for main grain storage hazard-moulds, a preparation method of an antifungal metabolite of the bacillus subtilis strain, and application of the bacillus subtilis strain and the antifungal metabolite in grain storage. The strain is bacillus subtilis LM3403, and has a preservation number of CGMCC No. 9377. The bacillus subtilis LM3403 has a better inhibiting effect for the main grain storage hazard-moulds such as fusarium graminearum, fusarium moniliforme, aspergillus flavus, aspergillus niger, aspergillus glaucus, aspergillus fumigatus, penicillium oxalicum, penicillium expansum and mucor mucedo, and is wide in antibacterial spectrum, thus having a giant application potential. The antifungal metabolite of the bacillus subtilis LM3403 has an obvious inhibiting effect for the growth of the main grain storage hazard-moulds. The storage test results prove that after the antifungal metabolite of the bacillus subtilis LM3403 is added into stored grains, the growth of the moulds can be effectively inhibited in a grain storage process, so that the grain storage safety is guaranteed.

Description

technical field [0001] The invention belongs to the technical field of grain storage, and in particular relates to a Bacillus strain broad-spectrum antagonizing major grain-harming molds, a preparation method for its antibacterial metabolite, and an application of the Bacillus strain and its antibacterial metabolite in grain storage. Background technique [0002] Compared with other countries in the world, my country's grain storage period is longer, with an average storage period of 3 years. In long-term storage, stored grains will inevitably be affected by external environmental factors and moisture migration in grain piles, resulting in mold growth, metabolism, and toxin production. The growth and metabolism of mold can decompose the nutrients in the grain and cause the quality of the grain to deteriorate to varying degrees. According to incomplete statistics, the annual loss caused by grain mildew in the world alone accounts for more than 3% of the total output. In my c...
