[0032] The invention also discloses a preparation method of the composite hydrogel, mixing and stirring nanoparticles and polysaccharides in an aqueous medium to obtain the composite hydrogel;

[0033] The nanoparticles are nano-silicon dioxide, nano-hydroxyapatite, particles containing nano-silicon dioxide or particles containing nano-hydroxyapatite.

[0034] In the preparation method of the present invention, nano particles, polysaccharide and aqueous medium are used as raw materials. The descriptions of the nanoparticles, polysaccharides and aqueous medium are the same as those of the above-mentioned technical solution, and will not be repeated here.

[0035] According to the present invention, the nano particles and the polysaccharide are mixed and stirred in the aqueous medium to obtain the composite hydrogel. The present invention has no special limitation on the mixing and stirring temperature, room temperature is enough, which is convenient for in-situ encapsulation o...

Embodiment 1

[0043] According to the literature ( W.; Fink, A.; Bohn, E.Controlled Growth of MonodisperseSilica Spheres in the Micron Size Range.J.Colloid Interface Sci.1968,26,62-69) prepared 40nm colloidal silica nanoparticles.

[0044] Stir and disperse colloidal silica nanoparticles in water to form a uniform dispersion system. An aqueous solution of carboxymethyl chitosan with a molecular weight of 200 kDa is prepared with water, mixed with bioactive nanoparticles of 40 nanometers, fully stirred, and a composite hydrogel is formed within 5 minutes. The final concentration of carboxymethyl chitosan was 80 mg / mL, and the final concentration of silica nanoparticles was 10 wt%.

[0045] Animal experiment: Take an adult 6-week-old Balb / c male mouse (average weight 20-25g) to remove a 1cm×1cm-sized skin tissue in the center of the back, and spread the above-prepared hydrogel evenly on the wound of the experimental group, every 2 days Apply once, 0.2mL each time. The mice were sacrificed...

Embodiment 2

[0048] Synthesis of 40 nm bioactive silica nanoparticles: In this example, colloidal silica nanoparticles with a particle size of 40 nm prepared by the sol-gel method were used as raw materials, and the preparation method refers to the literature described in Example 1. First, the colloidal silica nanoparticles are stirred and dispersed in water to form a uniform dispersion system. Then, the saturated aqueous solution of calcium hydroxide is added in the aqueous dispersion system of above-mentioned colloidal silicon dioxide nanoparticle, and wherein the mol ratio of silicon dioxide and calcium hydroxide is 50:1, the mass of water and silicon dioxide in the system The ratio is 50:1. The system was magnetically stirred at 25° C. for 8 hours, centrifuged, washed with water to remove unreacted calcium hydroxide, and dried to obtain biologically active nanoparticles with a particle size of about 40 nanometers.

Abstract

The invention relates to the field of hydrogel, and discloses a composite hydrogel as well as a preparation method and an application thereof. The composite hydrogel is formed by cross-linking polysaccharides and nanoparticles in an aqueous medium, wherein the nanoparticles are nano silicon dioxide, nano-hydroxyapatite, particles containing the nano silicon dioxide or particles containing the nano-hydroxyapatite. The composite hydrogel, which takes the nanoparticles as physical cross-linking points, can a non-covalent bond effect with chain segments of the polysaccharides, so that cross-linking with the chain segments of the polysaccharides is realized. Experimental results show that the composite hydrogel, by mixing the nanoparticles, is relatively good in effects of promoting cell proliferation and accelerating wound healing. Moreover, the composite hydrogel provided by the invention can be generated under gentle conditions; and the preparation method is simple.

Description

technical field [0001] The invention relates to the field of hydrogels, in particular to a composite hydrogel, its preparation method and application. Background technique [0002] Hydrogel is a new type of polymer material with a three-dimensional network structure, which uses an aqueous medium as a dispersion medium. [0003] The hydrogel has good permeability to low-molecular substances, high water content, fast swelling, good biocompatibility, and slow-release performance of drugs. Therefore, hydrogels can not only be developed as sensors, artificial muscles, cell immobilization and memory materials, but also can be used as drug carriers and tissue engineering matrix materials. [0004] Hydrogels are obtained by chemically or physically crosslinking water-soluble or hydrophilic polymers. These polymers can be divided into two categories according to their sources: natural and synthetic. Natural hydrophilic polymers include polysaccharides (starch, cellulose, alginic ac...
