Example Embodiment

[0032] The invention also discloses a method for preparing the composite hydrogel. The nano particles and polysaccharides are mixed and stirred in an aqueous medium to obtain the composite hydrogel;

[0033] The nanoparticles are nano-silica, nano-hydroxyapatite, particles containing nano-silica or particles containing nano-hydroxyapatite.

[0034] In the preparation method of the present invention, nanoparticles, polysaccharides and an aqueous medium are used as raw materials. The descriptions of the nanoparticles, polysaccharides and aqueous medium are the same as the above technical solutions, and will not be repeated here.

[0035] According to the present invention, the nanoparticles and polysaccharides are mixed and stirred in an aqueous medium to obtain a composite hydrogel. The invention has no special restrictions on the temperature of mixing and stirring, as long as room temperature is sufficient, which is convenient for in-situ encapsulation of biologically active substan...

[0042] Example 1

[0043] According to the literature ( W.; Fink, A.; Bohn, E. Controlled Growth of Monodisperse Silica Spheres in the Micron Size Range. J. Colloid Interface Sci. 1968, 26, 62-69) to prepare 40 nanometer colloidal silica nanoparticles.

[0044] The colloidal silica nanoparticles are stirred and dispersed in water to form a uniform dispersion system. An aqueous solution of carboxymethyl chitosan with a molecular weight of 200 kDa is prepared with water, mixed with 40 nanometer bioactive nanoparticles, fully stirred, and a composite hydrogel is formed in 5 minutes. The final concentration of carboxymethyl chitosan was 80 mg / mL, and the final concentration of silica nanoparticles was 10 wt%.

[0045] Animal experiment: Take adult 6-week-old Balb / c male mice (average weight 20-25g) to remove 1cm×1cm skin tissue in the center of the back, and apply the above-prepared hydrogel evenly on the wound of the experimental group, every 2 days Apply once, 0.2mL each time. Afte...

[0047] Example 2

[0048] Synthesis of 40 nanometer bioactive silica nanoparticles: In this embodiment, colloidal silica nanoparticles with a particle size of 40 nanometers prepared by the sol-gel method are used as raw materials, and the preparation method refers to the document described in Example 1. First, the colloidal silica nanoparticles are stirred and dispersed in water to form a uniform dispersion system. Then, a saturated aqueous solution of calcium hydroxide was added to the water dispersion system of colloidal silica nanoparticles, where the molar ratio of silica to calcium hydroxide was 50:1, and the mass of water and silica in the system The ratio is 50:1. After the system was magnetically stirred at 25°C for 8 hours, centrifuged and washed with water to remove unreacted calcium hydroxide. After drying, biologically active nanoparticles with a particle size of about 40 nanometers can be obtained.

Abstract

The invention relates to the field of hydrogel, and discloses a composite hydrogel as well as a preparation method and an application thereof. The composite hydrogel is formed by cross-linking polysaccharides and nanoparticles in an aqueous medium, wherein the nanoparticles are nano silicon dioxide, nano-hydroxyapatite, particles containing the nano silicon dioxide or particles containing the nano-hydroxyapatite. The composite hydrogel, which takes the nanoparticles as physical cross-linking points, can a non-covalent bond effect with chain segments of the polysaccharides, so that cross-linking with the chain segments of the polysaccharides is realized. Experimental results show that the composite hydrogel, by mixing the nanoparticles, is relatively good in effects of promoting cell proliferation and accelerating wound healing. Moreover, the composite hydrogel provided by the invention can be generated under gentle conditions; and the preparation method is simple.

Description

technical field [0001] The invention relates to the field of hydrogels, in particular to a composite hydrogel, its preparation method and application. Background technique [0002] Hydrogel is a new type of polymer material with a three-dimensional network structure, which uses an aqueous medium as a dispersion medium. [0003] The hydrogel has good permeability to low-molecular substances, high water content, fast swelling, good biocompatibility, and slow-release performance of drugs. Therefore, hydrogels can not only be developed as sensors, artificial muscles, cell immobilization and memory materials, but also can be used as drug carriers and tissue engineering matrix materials. [0004] Hydrogels are obtained by chemically or physically crosslinking water-soluble or hydrophilic polymers. These polymers can be divided into two categories according to their sources: natural and synthetic. Natural hydrophilic polymers include polysaccharides (starch, cellulose, alginic ac...
