Embodiment
[0203] , Extraction of RNA from paraffin samples
[0204] After extracting the RNA of the breast cancer sample, the sample A to be tested is obtained;
[0205] 1) Put the breast cancer sample into a 1.5ml centrifuge tube;
[0206] 2) Add 240ml PKD Buffer, shake and mix, and centrifuge at 11000g for 1min;
[0207] 3) Add 10ul proteinase K, pipette gently to mix;
[0208] 4) Incubate at 56°C for 15 minutes, then at 80°C for 15 minutes;
[0209] 5) Incubate on ice for 3 minutes, centrifuge at 20000g for 15 minutes;
[0210] 6) Transfer the upper liquid to a new 1.5ml centrifuge tube;
[0211] 7) Add 25ul of DNase Booster Buffer (about 1 / 10 of the liquid in the centrifuge tube), and 10ul of DNaseI stock solution, mix upside down and centrifuge briefly to mix the liquid on the tube wall into the bottom;
[0212] 8) Incubate at room temperature for 15 minutes;
[0213] 9) Add 500ul Buffer RBC to adjust the binding environment and mix well;
[0214] 10) Add 1200ul absolute eth...

Abstract
The invention discloses a breast cancer 21 gene detection kit and a detection method thereof. The kit comprises primers and probes for detecting genes ACTB, GAPDH, RPLPO, GUS, TFRC, CD68, BAG1, GSTM1,HER2, GRB7, MMP11, CTSL2, KI67, STK15, CCNB1, MYBL2, Survivin, ESR1, PGR, BCL2 and SCUBE2. Two pairs of primers and two specific probes are respectively designed for 21 genes in the kit, a primer andprobe liquid of each gene is placed in one reaction tube, a same buffer solution and a procedure are adopted for QPCR (Quantitative Polymerase Chain Reaction) amplification, the operation is simple and convenient to carry out, the detection time can be shortened, and clear and accurate results can be achieved; the kit is wide in application, and applicable to both scientific research and clinicaldetection and applicable to single or multiple samples, and rapid and accurate detection results can be achieved.
Description
technical field [0001] The invention relates to the field of gene technology, in particular to a breast cancer 21 gene detection kit and a detection method thereof. Background technique [0002] Breast cancer is one of the most common malignant tumors in women. In the past 20 years, the multidisciplinary comprehensive treatment model centered on surgery and assisted by chemotherapy, radiotherapy, and endocrine therapy has been greatly developed, which has significantly increased the recurrence and mortality of breast cancer. However, there is still a lack of effective methods that can accurately predict the recurrence risk of patients and give corresponding treatment. [0003] The breast cancer recurrence risk assessment system is based on RT-QPCR method to detect the expression of 16 tumor-related target genes in tumor tissue through precise calculation, that is, breast cancer 21 gene detection, and the breast cancer risk is obtained by calculating the amount of gene expres...
