Problems solved by technology

[0015] However, this description is problematic in more ways than one
First, no information is presented about the particular decomposition process
Also, no identification is given of what a sufficiently large amount would be
Thus, however, efficient and timely detection of Chlamydia appears to be difficult

Embodiment Construction

[0060] Tests were performed with Chlamydia trachomatis species that had been grown in a cell line of HeLa cells. The cell line of Hela cells is a cell type among immortal cell lines used in scientific research. Sample preparation in Figure 4 shown in . The starting composition contained A (the species in HeLa cells) in medium M. HeLa cells were lysed by sonication using three sonication periods (20 seconds each) at 70 W interrupted by 2 second rest periods using vortexing. This results in the protomers (EB) within which can be collected, while other cellular material is destroyed into cellular debris (D). After sonication, EB+D is thus present in the medium (M). The cellular material is then transferred to a centrifuge for separation of cellular debris (D) and protomers (EB). In this example, three subsequent steps (C1 , C2, C3) were performed using increasing spin rates, namely 500xg, 2500xg and 15,000xg. Thus, first the medium M is removed, then a part of the cell deb...

Abstract

The method for the diagnosis of a microbial infection in an organism makes use thereof that said microbial infection is at least partially present as elementary bodies in cellular material of the organism. The method comprises the steps of (1) providing a sample of cellular material from the organism; (2) processing the sample to obtain a test composition, that is enriched in elementary bodies, inso far as the sample contains any elementary bodies; and (3) Subjecting a volume of the test composition comprising at most a predetermined maximum of elementary bodies to a MALDI mass spectrometry method to identify presence of the microbial infection. The sample processing more particularly involves cell lysis and separation of elementary bodies from the lysed cell material. Subsequently, the elementary bodies (and any further material attached thereto) are contacted with a matrix material, that facilitates the MALDI mass spectrometry.

Description

[0001] field of invention [0002] The invention relates to a method for diagnosing a microbial infection in an organism, wherein the microbial infection is present at least partially in the cellular environment of the organism as inclusion bodies. [0003] The invention also relates to a system for diagnosis. [0004] Background of the invention [0005] Microbial infections lead to threats to the health of organisms such as mammals and more particularly humans. A variety of diagnostic methods are available to identify such infections. However, such identification can be laborious. Furthermore, microbial infections can be asymptomatic such that the infected host organism is unaware of infection. This allows the infection to spread. In addition, some microbial infections occur as intracellular infections. [0006] In a broad sense, certain classes of infections are at least partially present as inclusion bodies. Such inclusion bodies are nuclear or cytoplasmic aggregates ...
