Abstract

The invention relates to a fluorescently-labeled PD-L1 membrane surface expression cell model and an application thereof. The cell model is a eukaryon host cell which can express overall length sequence of PD-L1 maturation protein, and the eukaryon host cell is transfected to a 293T cell through a eukaryon fluorescence expression vector which is constructed; the eukaryon fluorescence expression vector realizes transient expression of the PD-L1 protein in the 293T cell, and the PD-L1 protein is shown in the surface of the 293T cell; and the cell model is applied to screening of polypeptide andantibodies, which can be combined with PD-L1. According to the fluorescently-labeled PD-L1 membrane surface expression cell model and the application thereof, a cattle Fc gamma receptor signal peptidesequence is replaced with signal peptide on an original PD-L1 gene, so that the PD-L1 can be efficiently anchored to a cell membrane, and dog PD-L1 maturation protein can be massively shown on the surface of the eukaryon cell membrane.

Description

technical field [0001] The invention relates to the technical field of genetic engineering, in particular to a PD-L1 membrane surface expression cell model with a fluorescent marker and an application thereof. Background technique [0002] Programmed death 1 (PD-L1, CD279) is a member of the CD28 family, expressed in B lymphocytes, T lymphocytes, NK cells, dendritic cells and monocytes, and its ligand PD -L1 (Programmed death ligand 1, CD274) is a member of the B7 family. As the main ligand of PD-L1, PD-L1 is mainly expressed in tumor cells, lymphocytes, and epithelial cells. Cancer is one of the common diseases causing death in dogs. The current treatment methods for dog cancer include surgery, chemotherapy and radiotherapy. In addition, immunotherapy has also attracted much attention, and some drugs targeting immune checkpoint molecules have been successfully developed. In many clinical trials, anti-PD-L1 antibodies have shown good anti-tumor activity, especially in the ...
