Embodiment 1

[0036] 1. Establishment of a mouse model of Schistosoma japonicum infection

[0037]Healthy SPF-grade 8-week-old BALB / c mice were divided into seven groups: normal control group, 3-day S. The 28-day infection group and the 42-day Schistosoma japonicum infection group had 10 mice in each group. The normal control group was not treated, and the mice in the infection group were infected according to the conventional method: take the positive snails and place them in dechlorinated water, release the cercariae under the light condition of 25°C-28°C for 1-2h, and dip the inoculation loop to pick up 30 cercariae on the water surface Collected on a coverslip. After the mice were anesthetized, the abdomen was fixed on the mouse board, and the abdomen was shaved and then moistened with dechlorinated water. The coverslips stained with cercariae were attached to the abdominal skin of the mice for 20 minutes to infect.

Abstract

The invention discloses a serum biomarker for early diagnosis of schistosomiasis japonica. The serum biomarker is phosphatidylcholine and / or palmitoyl choline. The sensitivity and specificity of the marker are both greater than 0.9, and the under-curve area (AUC) is greater than 0.9, which indicates that the two indexes have favorable predictability and can be used for early diagnosis of schistosomiasis japonicum katsurada. The invention further discloses a screening method of the serum biomarker for early diagnosis of schistosomiasis japonica based on metabonomics. A mouse schistosomiasis japonica model is constructed, and the metabonomics analysis is carried out on mouse serum by utilizing an ultra-high performance liquid chromatography-tandem mass spectrometry technology, and characteristic differential metabolites between a normal mouse and a schistosomiasis japonica infected mouse are found and analyzed, namely, the schistosomiasis japonica early diagnosis biomarker is obtained. The screening method has the characteristics of convenience and quickness, can accurately reflect the metabolic spectrum difference between a schistosomiasis japonicum infected mouse and a normal mouse, and is high in specificity.

Description

technical field [0001] The invention relates to the technical field of biomedicine, and more specifically, relates to a serum biomarker for early diagnosis of schistosomiasis japonicum, a screening method and application. Background technique [0002] Schistosoma japonicum is a zoonotic parasitic disease caused by Schistosoma japonicum that seriously endangers human health and hinders social and economic development. It is widely prevalent in Asia, especially in China, the Philippines and Indonesia, where more than 100 Ten thousand people are infected with the disease, and the threatened population is about 46 million people. In my country, Schistosoma japonicum is mainly prevalent in 12 provinces, municipalities and autonomous regions in the middle and lower reaches of the Yangtze River and its south. After more than 60 years of hard work, the prevention and control of schistosomiasis in my country has achieved remarkable results. Now the endemic areas are mainly distribut...
