Abstract

The invention discloses a male-promoting gene MmS for populus deltoides and application thereof, and belongs to the technical field of plant genetic engineering. The specific male-promoting gene MmS of the populus deltoides disclosed by the invention does not encode a protein, but rather as a miRNA sponge for adsorbing miRNA; and through reducing concentration of the adsorbed free miRNA, an expression level of a down-stream target gene is up-regulated. The MmS gene exists in a male plant of the populus deltoides in a form of hemizygote, and only exists on a Y chromatid; and an X chromatid doesnot contain the gene. The male-promoting gene makes a male flower of the male plant of the populus deltoides to be developed, and the male plant of the populus deltoides does not contain the gene, and therefore, a male flower organ of the male plant of the populus deltoides is not developed. A large amount of pollen can be produced after the male plant of the populus deltoides reaches sexual maturity, so that severe environmental pollution is caused; and the MmS gene is knocked out in the male plant of the populus deltoides, so that a new variety of the populus deltoides which cannot producethe pollen can be cultured, the gene also can be imported into the plant, the development of male flowers is promoted, and thus, the gene has an important application value in seed cultivation of thepopulus deltoides.
