The method according to the present invention comprises measuring the amount of a peptide as a biomarker contained in a biological sample with the use of LC-MS and detecting a specific disease on thebasis of the measurement result, wherein pretreatment steps to be conducted before the measurement with the use of LC-MS involve: a step for adding to the biological sample a stable isotope reagent, said reagent being obtained by labeling a peptide with a stable isotope, and trifluoroacetic acid to give a mixed liquid sample; a step for boiling the mixed liquid sample; a step for pouring the boiled mixed liquid sample into a solid phase extraction column and allowing the solid phase extraction column to hold the peptide; and a step for passing a water-soluble organic solvent through the solidphase extraction column and thus eluting the peptide held by the solid phase extraction column.
