Problems solved by technology

However, the cross-linked PNIPAAm hydrogel itself cannot be degraded in vivo, which greatly limits its application.
Currently, there is no report on the preparation of pH-sensitive biodegradable PVME / CMCS hydrogels using semi-IPN technology.

Embodiment 1

[0014] Embodiment 1: 0.1M carboxymethyl chitosan and 0.1M polymethyl vinyl ether were mixed in a volume ratio of 15:15, magnetically stirred for 120 minutes, fully mixed, and 0.75ml of 1wt% pentadiene was added dropwise Aldehyde, continue to stir and react for 1h, and after standing for 5 minutes, cast a film in a polytetrafluoroethylene mold, and volatilize to a constant weight at room temperature. The resulting hydrogel has an equilibrium swelling degree of 1395% at pH=1.0 at 20°C, an equilibrium swelling degree of 185% at pH=3.0, a 544% equilibrium swelling degree at pH=4.4, and an equilibrium swelling degree of at pH=7.4. The equilibrium swelling degree is 1622%.

Embodiment 2

[0015] Example two: 0.1M carboxymethyl chitosan and 0.1M polymethyl vinyl ether were mixed in a volume ratio of 12:18, magnetically stirred for 120 minutes, fully mixed, and 0.6ml of 1wt% pentadiene was added dropwise Aldehyde, continue to stir and react for 1h, and after standing for 5 minutes, cast a film in a polytetrafluoroethylene mold, and volatilize to a constant weight at room temperature. The equilibrium swelling degrees of the obtained hydrogel at 20°C and pH=1.0, 3.0, 4.4 and 7.4 were 630%, 322%, 129% and 409%, respectively.

Embodiment 3

[0016] Embodiment three: 0.1M carboxymethyl chitosan and 0.1M polymethyl vinyl ether were mixed in a ratio of 18:12 in molar ratio, magnetically stirred for 120 minutes to make it fully mixed, and 0.9ml of 1wt% pentamethylenediene was added dropwise Aldehyde, continue to stir and react for 1h, and after standing for 5 minutes, cast a film in a polytetrafluoroethylene mold, and volatilize to a constant weight at room temperature. The equilibrium swelling degrees of the obtained hydrogel at 20°C and pH=1.0, 3.0, 4.4 and 7.4 were 1052%, 93%, 330% and 1458%, respectively.

Abstract

A pH sensitive hydrogel and its production are disclosed. The hydrogel consists of carboxy-methyl chitose, poly-methyl-vinyl ether, cross-linking agent glutaric dialdehyde and water. The proportion of carboxy-methyl chitose: poly-methyl-vinyl ether is 1:0.25-1.5 mol; proportion of cross-linking glutaric dialdehyde: carboxy-methyl chitose is 1:10-30 mol, residue is water. The production is carried out by agitating, dissolving carboxy-methyl chitose and poly-methyl-vinyl ether into deionized water in proportion of 1:0.25-1.5 mol, mixing, controlling concentration of them between 0.05-0.1 M, dripping glutaric dialdehyde into mixed solution in proportion of 1:10-30 mol, agitating while reacting for 30-60 mins, laying aside, pouring it into mould, drying at room temperature and weighing to obtain semi-penetrating network pH sensitive hydro-gel. It has excellent sensitivity, adhesion, flexibility and degradability. It can be used as medicine slow-releasing carrier.

Description

technical field [0001] The invention relates to a pH-sensitive hydrogel and a preparation method thereof, in particular to a pH-sensitive hydrogel with a semi-interpenetrating network structure and a preparation method thereof. Background technique [0002] Polymer gel refers to a macromolecular network filled with liquid, which is composed of polymers with a network structure and solvents. It is a three-dimensional network between liquid and solid, which has both liquid-like mobility and fluidity, and solid-like shape stability. When the macromolecular skeleton is hydrophilic, the liquid it fills can be water or an aqueous solution. This gel is called a hydrophilic gel, or hydrogel for short. [0003] Smart hydrogels are a class of hydrogels that respond to subtle changes in the external environment (such as temperature, pH, ionic strength, light, and specific chemical substances, etc.). pH-sensitive hydrogel is a polymer hydrogel whose volume changes with the pH value of...
