Embodiment Construction

[0029] A: Measuring the effect of different temperatures on the heart rate of zebrafish:

[0030] 1. Cultivate zebrafish embryos, connect the high-speed CCD to the image output terminal of the microscope, open the computer image acquisition software, and test whether it can work normally.

[0031] 2. Set the experimental environment temperature to 26°C.

[0032] 3. Put a little Hower's solution on the cover slip, and put the zebrafish under study into the Hower's solution on the cover slip with a pipette.

[0033] 4. Put the above-mentioned cover glass on the stage to form a relatively clear image of the heart part to be studied.

[0034] 5. Turn on the high-speed CCD, collect images and name the obtained video file fishdata.avi.

[0035] 6. Select a frame of image as the reference frame according to the above-mentioned calculation formula of corr (i, j), such as making i=66, n from 0 to 201600, and the value of j from 0 to 500 to obtain a series of heartbeat data. The data...

Abstract

The invention belongs to the technical field of biology measurement, in particular to a method of measuring the heart rate of the zebra fish by utilizing a high-speed CCD. The method comprises that the heart part of the zebra fish is imaged and a video file is collected by the high-speed CCD, then the correlated image process to the obtained video file is carried out, namely an interested image with one frame is chosen as a reference image, the other images and the reference image are calculated in a correlation way, then the data of the heart rate of the zebra fish is obtained. The method of the invention belongs to a non-contact measurement and improves the reliability of the research data without any damage to the creature.

Description

technical field [0001] The invention belongs to the technical field of biological measurement, in particular to a method for measuring the heart rate of zebrafish. Background technique [0002] Zebrafish is widely used in various biological experiments because of its relatively fast embryonic development (as shown in Figure 1). Its heart rate measurement is extremely important in biological experiments and new drug development. Internationally, green fluorescent protein or acupuncture is usually used to measure heart rate at present. These two methods are very cumbersome, and they are also prone to interference, resulting in inaccurate heart rate data. [0003] 1. GFP Method for Heart Rate Measurement: [0004] The method is to allow fish embryos to absorb green fluorescent protein through transgenic methods during embryonic development. This protein substance will stay in the heart of the fish and emit fluorescence. During the heart rate process, the green fluorescent prot...
