Embodiment 1

[0013] (1) Raw material preparation: prepare clover seeds, Glomus mosseae strains, sieve river sand and use 10 4 GyCo 60 Sterilization by gamma-ray irradiation.

[0014] (2) Preparation of inoculant: use river sand as the culture substrate and clover as the host plant, spread river sand, AM bacteria, river sand, clover seeds, and river sand in the cultivation pot from bottom to top in sequence, pour water thoroughly and place Cultivate in a culture room; after 120 days, all the aboveground parts of the clover are cut off, and the river sand containing fungal spores, extraroot hyphae, and infected clover root system is mixed and mixed to prepare a mycorrhizal fungus inoculant. The number of spores in the inoculum was determined by wet sieve decantation, and it was found that there were about 100 AM fungal spores per gram of inoculum.

[0015] (3) Seedbed preparation: use 10 4 GyCo 60 Sterilize by γ-ray irradiation; lay seedbeds in the order of "soil--inoculant--soil" from b...

Embodiment 2

[0019] The difference between this example and Example 1 is that the soil used in the seedbed preparation stage is not sterilized.

[0020] According to the statistics of the test, the infection rate and yield of Example 1 and Example 2 are significantly improved compared with the control without AM mycorrhizal fungus inoculation, see Table 1 for details. And it has different effects on the output of miniature tubers of different sizes, wherein embodiment 1 significantly increases the output of more than 4 grams of larger tubers, and embodiment 2 has the most obvious effect on increasing the production of medium miniature tubers with an individual weight of 2-4 grams, specifically see the table 2.

The number of spores in the inoculum was measured by wet sieve decantation method, and it was found that there were about 50 AM fungal spores per gram of inoculum.

Abstract

The invention relates to an application technology of AM mycorrhizal fungi in virus-free potato test tube plantlet transplanting. The technology comprises the following steps of: preparing an inoculant containing fungal spores, external mycelia, an infected clover root system and a river sand mixture by taking clovers as host plants; then preparing a transplanting substrate; and adding the inoculant to an interlayer of the substrate with a proportion of 3,000-5,000 spores per kilogram of soil. By adopting the method to transplant, the output of minituber virus-free potatoes can be increased by more than 25 percent.

Description

1. Technical field [0001] The invention relates to an application method of biological strains in agricultural production, in particular to an application method of AM mycorrhizal fungi in transplanting virus-free potato test-tube seedlings. 2. Background technology [0002] Potato is an important grain, vegetable, feed and industrial raw material. It has the characteristics of high yield, strong adaptability and rich nutrition. It has become the fourth largest food crop in the world. my country is the largest potato planting and producing country in the world, and its planting area ranks first in the world. With the application of tissue culture methods in production, the method of using tissue culture technology to produce virus-free seedlings to increase crop yields has been widely used in horticultural crops such as vegetables and flowers. In recent years, the technology of virus-free potato seedlings has been rapidly promoted, and has played an active role in increasin...
