Embodiment Construction

[0056] In order to make the technical means, creative features, goals and effects of the present invention easy to understand, the present invention will be further described below in conjunction with specific illustrations.

[0057] The specific operation process of the ship ballast water membrane concentration automatic counting method is as follows:

[0058] Taking the detection of ship ballast water samples as an example, 1m of ship ballast water samples were taken on site 3 → Filtration on site to obtain a concentrated water sample of about 100ml → Vacuum filtration of the concentrated water sample in the laboratory → Microscopic imaging of the filter membrane → Stitching images at the workstation → Automatic counting by software → Manual counting by looking at pictures.

[0059] Since the amount of living microorganisms ≥ 50um in ship's ballast water is very small, the concentration in seawater is usually 100 / m 3 , so each sample must be collected not less than 1m 3 ba...

Abstract

The invention discloses a method for automatically counting by concentrating through a filter membrane, which comprises the following steps: sampling ship ballast water on site; adopting a bio-filter, and filtering on site to obtain a concentrated water sample; performing vacuum filtration on the concentrated water sample by adopting a filter, wherein the thickness of the filter membrane of the filter is 10 mu m, the apertures are 10 mu m and the dimension of the apertures is consistent, and the filtering time is about 15 minutes; after filtering, arranging the filter membrane under a microscope for microscopic imaging; splicing images of a local area photographed by the microscope by using a work station and software for counting; automatically counting by using the software, automatically counting the number of objects with the dimension more than or equal to 50 mu m in the spliced images by using the automatically image counting function, and comparing the images with biological indicators of living bodies of the ballast water; and after automatically counting by using the software, counting the number of living beings through naked eyes by a detector when the living bodies exceed the indicators. The method has the advantages of higher accuracy, short time and low workload.

Description

technical field [0001] The invention relates to an automatic counting method for filter membrane concentration, in particular to an automatic counting method for ship ballast water filter membrane concentration, which belongs to the technical field of environmental microorganism monitoring. Background technique [0002] Ships transport more than 80% of the world's merchandise traffic, and approximately 10 billion tons of ballast water are transferred globally each year. It is estimated that more than 3,000 species of animals and plants migrate around the world through ballast water every day. The damage of marine invasive species caused by ships' ballast water to the marine environment has been recognized by the Global Environment Fund as one of the four major threats to the ocean (Bai Mindong et al., 2005). [0003] Due to the seriousness of the ballast water problem, all countries have paid more attention to it, and have invested a lot of manpower and material resources i...
