Abstract

The invention relates to an cell in-situ active deformation measurement method, which comprises the following measurement steps: (1) dividing square sampling grids on a cell-base fluorescent reference image and a corresponding deformation image; (2) determining the sizes of a reference subarea and a searching area; (3) establishing four global summation table; (4) calculating the cross correlation coefficient between the image reference subarea and the corresponding searching area, which take a sampling point as a center, by recurrence; (5) quickly calculating a zero-mean normalization related coefficient matrix by looking up the summation table according to the result of the step (4), and determining the overall pixel displacement of the sampling point; (6), performing subpixel interpolation by using a gradient method; and (7) recurring and calculating the displacement of the sampling point to obtain an accurate displacement field of a whole deformed image. The method has the advantages of accurate and high-efficiency measurement and high time-space resolution.
