Abstract

The invention aims to provide a microbial strain arthrobacter nitroguajacolicus, slant medium, fermentation medium and application thereof. The 16S rDNA sequence of the strain is shown as SEQ ID No. 1. According to the formula, the slant medium comprises 0.2 to 2.0 weight percent of carbon source, 0.2 to 2.0 weight percent of nitrogen source, 0.01 to 0.5 weight percent of trace metal based on the amount of the metal salt, and 1.0 to 3.0 weight percent of agar powder, wherein the pH is 6.4 to 7.5. According to the formula, the fermentation medium comprises 0.5 to 3.0 weight percent of carbon source, 0.2 to 3.0 weight percent of nitrogen source, 0.01 to 0.5 weight percent of trace metal ion based on the amount of the metal salt, 0.01 to 0.1 weight percent of cysteine hydrochloride or cysteine and 0.01 to 0.1 weight percent of organic nitrile, wherein the pH is 6.4 to 7.5. The strain can be used for producing nitrilase; and the nitrilase produced by using the strain can catalyze 3-hydroxybutyronitrile so as to produce 3-hydroxybutyric acid. The 3-hydroxybutyric acid prepared by using the method has the advantages of cheap and readily available raw material, less hydrolysis byproduct, mild reaction condition and the like; and thus the microbial strain has high industrial production potential.
