Embodiment Construction

[0124] like image 3 As shown, a mixture of a lipid-based imaging agent (MP1950, University of Virginia) and a polymer-based thick-shell microcapsule agent (IJ05232 with a diameter of 3.26 μm, Philips Research) was first mixed and injected into the tank. Position the fan-shaped ultrasound probe S3 connected to the Sonos 7500 ultrasound scanner vertically and place a double layer of sound-absorbing material on the bottom. The ultrasound scanner is operated using a contrast mode consisting of a low MI monitor mode for continuous contrast imaging and a pulsed mode (much higher MI) for contrast agent destruction. like Figure 4a As shown, in the monitoring mode with a very low transmit power of -22dB (corresponding to the "degraded" MI=0.1 on the display or the actual MI~0.16 at 3cm in water), only the imaging agent MP1950 microbubbles are seen with a high reception of 80dB gain. In Figure 4b, microbubbles of imaging agent MP1950 and polymer agent IJ05232 were observed in pulse...

Abstract

Described is a method for the delivery of a therapeutic agent to a mammal, particularly a human person, comprising (i) the administration of ultrasound particles that undergo a physical change when subjected to ultrasound; (ii) the administration of the therapeutic agent in an ultrasound dosage form allowing release of the therapeutic agent to be affected by the application of ultrasound, (iii) the application of ultrasound so as to affect a physical change in at least some of the ultrasound particles, and (iv) the application of ultrasound so as to stimulate release of the therapeutic agent. According to the invention the ultrasound particles are subjected to detection of their acoustic responses via echoes and / or emissions. The information thus obtained is processed and the processed information is used to adapt or adjust the ultrasound-mediated delivery of the therapeutic agent. This can be carried out using a single set of particles (that serve both as ultrasound imaging tracers and as drug delivery vehicles) or by applying separate sets of particles.

Description

technical field [0001] The present invention relates to administration and delivery of therapeutic agents mediated through the use of ultrasound. Background technique [0002] Therapeutic use of ultrasound-activated medicated particles is an emerging technology with potential for various clinical applications. In the fields of ultrasound contrast imaging and drug delivery, both conventional (untargeted) and targeted contrast agents can be used. For non-site-specific contrast agents, targeted drug delivery can be achieved by precisely focusing ultrasound energy at the region of interest. Ultrasound contrast agents, typically between 0.1 and 8 μm in diameter, increase the echo amplitude from the blood pool and enable detection of blood vessels even as small as capillaries. [0003] The rationale for using ultrasound-mediated drug delivery is based on the need for site-specific applications, such as organ or tissue-specific, tumor-specific or clot-specific drug therapy. Drug...
