Abstract

The invention which belongs to the technical field of biological heredity relates to a nano-particle molecule haplotype typing technique. Isolation and amplification of chromatids of a target DNA fragment can be realized through treating a genomic DNA of a sample to be detected as a template, and processing specific forward allele primers corresponding to SNP alleles of the target DNA fragment and a universal reverse primer with a long fragment AuNP ASPCR. According to the invention, the AuNP is innovatively added to an ASPCR system, so the specificity of haplotype typing is effectively improved. Obtained haploid PCR products can be directly used in SNP genotyping, for example, a haplotype of individuals of a diploid to be detected can be obtained through a TaqMan probe and sequencing. The other haplotype of the sample to be detected can be obtained through the same technique or can be conjectured by genotype.
