[0244] Mitsugumin 29 (MG29): A protein related to muscle-specific synaptophysin was isolated. The skeletal muscle triad is composed of a single depressed portion of the plasma membrane (transverse or T-tubules) trapped in the cytoplasm that is juxtaposed with two portions of the terminal pool of the sarcoplasmic reticulum (SR). Given the importance of the triplet in inducing muscle contraction, an antibody library was screened by immunostaining for novel proteins localized in the triplet, identifying regulators of excitation-contraction (E-C) coupling and Ca in skeletal muscle 2+ Other proteins that regulate other aspects are not surprising. One of the most important proteins identified during the screening of this library was a new member of the synaptophysin family of transmembrane proteins, mitsugumin 29 (MG29).

Abstract

Disclosed herein are compositions and methods for treatment of muscle dysfunction, including diabetes. In addition, the invention relates to therapeutic compositions comprising nucleotides and/or polypeptides of the invention in combination with a pharmaceutically acceptable carrier, wherein the composition facilitates the treatment of skeletal musle disorders. Moreover, the invention relates to the treatment and/or prevention of pathological conditions associated with altered intracellular Ca2+ reulgation and disrupted membrane structure that occurs when the expression levels of MG29 are reduced.
