AI Technical Summary

Benefits of technology

This patented technology allows for better control over blood flow during medical procedures by delivering high levels of dissolved gases (O2) into specific areas where they are needed without losing their ability to transport enough oxygen or create harmful substances like nitrogen oxides). It also helps improve brain function after surgery while still providing effective treatments against neurologically disorders such as stroke caused by strokes.

Problems solved by technology

The technical problem addressed in this patents relates to developing a safer and easier alternative route towards treatments like hyperoxygenemia. Current methods involve administering oxo carriers alone, making them difficult to control over dosage levels within specific areas inside the patient's body. There is thus a desire to develop improved ways to introduce these substances safely throughout the entire body while minimizing side effects associated with traditional routes.

Embodiment 1

[0039] Example 1 Preparation of Oxyflurolipid Microbubbles

[0040] 1) Weigh 50 mg DSPC and 20 mg DPPE-MPEG5000 into a capsule bottle with a precision weighing device, add 50 μl glycerin and 450 μl PBS; 2) Heat in a constant temperature water bath at 55°C for 30 minutes to completely dissolve DSPC and DPPE-MPEG5000 in the solution; 3) Seal the capsule bottle with a parafilm; 4) Use a syringe to extract 3 volumes of oxygen and 1 volume of perfluoropropane, and use a three-way tube to mix oxygen and perfluoropropane under negative pressure; 5) Use a three-way tube, Replace the air in the capsule bottle with mixed gas under negative pressure to reach a saturated state; 6) After mechanically shaking with an amalgam capsule blender, a white emulsion-like lipid microbubble suspension is obtained, namely fluoride lipid Microbubbles, stored in a refrigerator at 4°C.

Embodiment 2

[0041] 2) Example 2 Preparation of Oxyflurolipid Microbubbles

[0042] 1) Weigh 50 mg DSPC and 20 mg DPPE-MPEG5000 into a capsule bottle with a precision weighing device, add 50 μl glycerin and 450 μl PBS; 2) Heat in a constant temperature water bath at 55°C for 30 minutes to completely dissolve DSPC and DPPE-MPEG5000 in the solution; 3) Seal the capsule bottle with a parafilm; 4) extract 4 volumes of oxygen and 1 volume of perfluoropropane with a syringe, and use a three-way tube to mix oxygen and perfluoropropane under negative pressure conditions; 5) use a three-way tube, Under negative pressure conditions, the air in the capsule bottle is replaced with a mixed gas to reach a saturated state; 6) After mechanically shaking with an amalgam capsule blender, a white emulsion-like lipid microbubble suspension is obtained, namely the oxyfluoro lipid Microbubbles, stored in a refrigerator at 4°C.

Embodiment 3

[0043] Example 3 Preparation of Oxyflurolipid Microbubbles

[0044] 1) Weigh 50 mg DSPC and 20 mg DPPE-MPEG5000 into a capsule bottle with a precision weighing device, add 50 μl glycerin and 450 μl PBS; 2) Heat in a constant temperature water bath at 55°C for 30 minutes to completely dissolve DSPC and DPPE-MPEG5000 in the solution; 3) Seal the capsule bottle with a sealing film; 4) Use a syringe to extract 5 volumes of oxygen and 1 volume of perfluoropropane respectively, and use a three-way tube to mix oxygen and perfluoropropane under negative pressure; 5) Use a three-way tube, Under negative pressure conditions, the air in the capsule bottle is replaced with a mixed gas to reach a saturated state; 6) After mechanically shaking with an amalgam capsule blender, a white emulsion-like lipid microbubble suspension is obtained, namely the oxyfluoro lipid Microbubbles (MB(5:1)), stored in a refrigerator at 4°C.

Abstract

The invention discloses an oxygen-fluorine liposome microbubble therapeutic agent which consists of microbubbles, and oxygen and at least one perfluocarbon existing in the microbubbles, wherein the gas nucleus takes oxygen as a principle component and takes perfluocarbon as an auxiliary component, and the microbubble mainly consists of phospholipid and polyethylene glycol derivative. According to the oxygen-fluorine liposome microbubble therapeutic agent, oxygen can be controlled to be released in a human body through veins delivery and targeted hypoxia tissue under ultrasound wave irradiation, the hypoxia disease is treated, and the therapeutic agent serves as a contrast agent used for ultrasonic enhanced imaging. Therefore, the microbubble is a developing agent and an oxygenating therapeutic agent.
