Abstract

The invention relates to the field of protection of plant germplasm resources, and in particular provides a method for ultra-low temperature preservation and grafting resurrection of dormant buds of mulberry trees, including 1) obtaining dormant branches; 2) cutting into dormant buds; 3) drying; 4) sealing; 5) Gradient cooling; 6) Liquid nitrogen preservation; (7) Thawing; 8) Rehumidification; 9) Grafting resurrection. The method of the invention is a method for ultra-low temperature preservation and grafting resurrection of dormant buds of mulberry trees by using a drying-gradient cooling and freezing method. The dormant buds of mulberry trees are dried to a water content of 20%-40%, and a higher survival rate is obtained through steps such as slow and step-by-step cooling treatment, freezing storage, thawing, rehumidification, and grafting and resurrection. By adopting the method provided by the invention, the average survival rate of dormant buds of mulberry trees after cryopreservation can reach up to 95%. The cryopreservation method for mulberry dormant bud segments provided by the invention has simple procedure, easy operation and good freezing effect, and is an effective way for long-term preservation of mulberry germplasm resources in vitro.
