Abstract

The invention relates to a label-free rapid optical detection method of three-dimensional distribution of age spots. The label-free rapid optical detection method is characterized by comprising the following steps: (1) firstly freezing to-be-detected tissues or organs in a low-temperature environment to ensure that the tissues or organs are hardened; (2) fixing the frozen tissues or organs into a low-temperature environment in which the tissues can be mechanically processed, and performing mechanical processing to obtain tissue slices or flat sections; (3) detecting an auto-fluorescence signal of the tissue slices or the flat sections by using a photoelectric detection method to obtain information about the position distribution of the age spots; and (4) repeating the steps (2) and (3) until data of the whole tissues or organs are completely acquired, thereby obtaining information about the three-dimensional distribution of the age spots. Compared with a conventional method, the method disclosed by the invention can be used for significantly shortening the time in a whole process from preparation of samples to acquisition of age spot distribution information through imaging; and complete three-dimensional information of age spot distribution can be acquired by using the method.
