Embodiment Construction

[0011] The technical solution of the present invention will be further described below in combination with experiments.

[0012] Experiment 1: Construction of JurkatHIV-1 latently infected cell line and the activation effect of tannic acid in this system.

[0013] 1. Experimental method:

[0014] 1) Take the well-grown human epi-renal cell line 239T cells and inoculate them in a 10cm flat-bottomed plate. The medium used is a complete medium: high-glucose DMEM, 10% fetal bovine serum and 1% double antibody, and the culture condition is 5% carbon dioxide, 37°C;

[0015] 2) After 24 hours of adherence, the transfection pNL4-3-transfection medium is a complete medium and VSV-G, as in 1) and continued to culture;

[0016] 3) After 48 hours, collect the supernatant, use PEG-6000 to concentrate the virus particles, and use the ELISAP24 kit to detect virus P24;

[0017] 4) After counting well-grown Jurkat cells, infect with the virus obtained in 3), the virus dosage is about 10ng...

Abstract

The invention discloses application of tannic acid serving as an HIV-1 latent infection activator. A Jurkat latent infection cell line of HIV-1 with an FGP reporter gene which is integrated is applied, it is proved that the tannic acid has the function of activating HIV-1 latency by conducting high throughput screening on patent medicine storage, and GFP expression quantity in a screening system is increased. It is further proved through the experiments in multiple HIV-1 latent infection cell model cloned strains such as J-Lat that the drug indeed has the good function of activating the HIV-1 latency. It is proved through the experiments that EC50 of the tannic acid in J-Lat cells is 46.63 microns, and a good activation effect of the HIV-1 latent infection is achieved, and the tannic acid has important research and development value and development significance in the aspect of activating the HIV-1 latent infection.

Description

technical field [0001] The invention relates to a new application of the compound, in particular to the application of tannic acid as an activator of HIV-1 latent infection. Background technique [0002] HIV-1 virus is a lentivirus that infects the cells of the human immune system. It attacks human T lymphocytes, destroys the human immune system, and eventually leads to the generation of acquired immunodeficiency syndrome AIDS. The virus was first discovered in 1981. For more than 30 years, researchers have been working on the control and elimination of the virus, so as to achieve the cure of AIDS. The current conventional treatment method is called HAART for short, highly effective antiretroviral therapy, which can effectively inhibit the replication of the virus, thereby controlling the viral load in the patient's peripheral blood to a low level. However, the current general treatment methods can only control the virus to a certain extent, and cannot effectively remove it...
