Embodiment Construction

[0026] The present invention will be further described below in conjunction with accompanying drawing:

[0027] 1. Using a home-style sheet cutting machine as the main tool for forming cellophane chips, designed and produced two-dimensional tiny paper chips (such as diameter: 3.0mm; thickness: 0.5mm), this paper chip does not require special modification or activation step;

[0028] 2. Sensitivity verification of mercury ion detection by fluorescence energy resonance transfer on a paper chip, the specific steps are:

[0029] (1) Take the sterilized paper chip and add 4.0 μL graphene oxide dropwise, and let it dry naturally;

[0030] (2) Add 2.0 μL Cy3-labeled mercury ion aptamer dropwise to the above paper chip; place it in a dark place until there is no obvious liquid accumulation on its surface;

[0031] (3) During the dark treatment in step (2), add 3.0 μL of mercury ion solution with a concentration gradient (gradient: 0.20.51.010.050.0200.0500.01000.0 μM) into the centr...

Abstract

The invention belongs to the technical field of paper chip micro fluidics, and particularly discloses a method for detecting a mercury ion on the basis of fluorescence resonance energy transfer of a paper chip. According to the method disclosed by the invention, the mercury ion of heavy metal is detected by taking a glass fiber chip as a carrier and by utilizing a method of the fluorescence resonance energy transfer. According to property that a Cy3 fluorescence dye labeled Hg<2+> aptamer (Cy3-Hg<2+>-aptamer) can be specifically bound with Hg<2+> to form a T-Hg<2+>-T structure, by taking the Cy3-Hg<2+>-aptamer as a fluorescence reporter molecule and graphene oxide as a fluorescence quenching agent, Hg<2+> is rapidly detected directly at low cost. The paper chip is designed and cut by a common household cutting machine, and does not need to be further modified or activated; according to the method disclosed by the invention, a fluorescence inverse microscope is also served as an excitation light source to provide excitation light for acquisition of a fluorescence picture, and finally, the acquired fluorescence picture is recorded by a digital camera, therefore a potential requirement is provided for disease diagnosis and biological analysis.

Description

technical field [0001] The invention belongs to the technical field of paper chip microfluidics, and in particular relates to a method for detecting mercury ions. Background technique [0002] Mercury is a toxicant that can accumulate in organisms and is easily absorbed through the skin as well as the respiratory and digestive tracts. Minamata disease is a type of mercury poisoning. Mercury damages central nervous tissue and has adverse effects on the mouth, mucous membranes and teeth. Prolonged exposure to high levels of mercury can cause brain damage and death. Despite mercury's high boiling point, mercury vapor saturated at room temperature is several times the poisoning dose. It is a heavy metal ion that is extremely toxic to the human body. [0003] As a two-dimensional material, paper has the advantages of good water absorption, large specific surface area, environmental protection, and good biocompatibility. In addition, because of its easy processing and formabil...
