Problems solved by technology

And seed purity is the core indicator of seed quality, due to the complexity of seed breeding, limited parental resources and new hybrid F 1 The continuous increase of the number of false hybrids formed in the seed production process will cause heavy losses to cucumber production and farmers' economic income
Traditional methods for testing seed purity mainly include identification of seed morphology and phenotypic identification of field plots, which are time-consuming and labor-intensive and are easily affected by the environment and human factors, and cannot effectively distinguish hybrids with close genetic relationships (Salgado et al, 2006) ; The reliability of the results of isoenzyme identification is poor, and it is greatly affected by the environment

Embodiment Construction

[0029]As we all know, most of the inheritance of plant organelles is maternal inheritance, that is, mitochondria follow the mother cell and enter the fertilized egg. The relevant research on cucumber shows that its mitochondrial gene is inherited from the paternal line, that is, the mitochondria follow the sperm into the fertilized egg. The present invention is based on the patrilineal inheritance characteristic of cucumber mitochondria, thereby confirming the feasibility of the cucumber mitochondrial genome SSR marker in the detection of seed purity.

Abstract

The invention discloses a method for identifying cucumber hybrid purity based on cucumber mitochondrial paternal inheritance characteristics by using a mitochondrial genome SSR marker technology, relates to cucumber mitochondrial genome SSR marker development as well as methods for respectively performing polymorphism screening on cucumber varieties of different ecotypes by using the developed cucumber mitochondrial marker and performing purity detection on cucumber F1 seeds by utilizing a polymorphism marker, belonging to the field of biotechnology breeding. According to the invention, the purity of cucumber hybrid first-generation seeds is detected by using a cucumber mitochondrial genome sequence development SSR marker, and false hybrid seeds and selfing seeds from other pollen sources on maternal cucumber plants are expected to be distinguished; relative to research in variety identification aspect based on a nuclear genome SSR marker technology, a novel detection method is provided by mainly utilizing the cucumber mitochondrial paternal inheritance characteristics, and a technical preparation is made for tracing paternal / maternal mitochondrial genomes in the hybridization process. The method also can be applied to detection of hybrid first-generation seeds of other paternal plant materials.

Description

1. Technical field [0001] The invention discloses the development of cucumber mitochondrial genome SSR markers and its application in seed purity identification, which belongs to the field of biotechnology breeding, and relates to the development of cucumber mitochondrial genome SSR markers, and uses the developed cucumber mitochondrial markers to screen polymorphisms of cucumber varieties of different ecotypes. , and the use of polymorphic markers for cucumber F 1 Seeds are tested for purity. 2. Technical background [0002] Cucumber (Cucumis sativus L.) is an annual herbaceous plant belonging to Cucurbitaceae. As one of the ten most important vegetable crops in the world, cucumber is also one of the main vegetable crops in my country. It is generally believed that cucumbers originated in India, and China is one of the secondary origin centers of cucumbers, which can be divided into six ecological types: North China type, South China type, European greenhouse type, Europe...
