The invention discloses a polypeptide for distinguishing and detecting a PCV2 field virus antibody. The polypeptide comprises one or two polypeptides with polypeptide sequences as shown in SEQ ID NO.1 to SEQ ID NO. 4. The invention also discloses preparation methods for the above-mentioned polypeptide, and a coupled polypeptide and a polypeptide of a polysaccharide carrier, and discloses an ELISAdetection kit containing the polypeptide and a preparation method thereof. The ELISA detection kit for distinguishing and detecting the PCV2 field-virus antibody and the vaccine antibody can judge whether a to-be-detected pig farm has a field circovirus infection risk or not through serologic detection; if a weaned piglet is detected that a PCV2 antibody is positive before immunization with a PCV2 vaccine, and the level of the PCV2 antibody is gradually decreased and the level of the a vaccine antibody is gradually increased after immunization with the PCV2 vaccine, the weaned piglet is indicated have a circovirus infection risk, or a sow is indicated to be infected by field circovirus which is transmitted to the piglet through a maternal antibody.
