Abstract

The invention discloses application of integrating ginkgo biloba leaf extract and donepezil to preparation of a drug for controlling Alzheimer's disease. Through a great number of experiments for screening, a comprehensive multi-index comprehensive index method and PCA are adopted for comparing ginkgo biloba leaf extract and donepezil single-use value with a ginkgo biloba extract-donepezil total effect value, so that the two medicines have a dose-effect relationship when the integrated dose is 0.5-1 time of that of clinical dose, and the treatment effect is optimal when the dose of the two medicines is one time of the clinical dose; when the dose is 2-6 times of that of the clinical dose, the treatment effect is weaker than that of the clinical dose, and is obviously superior to that of single-use dose of the two medicines; when the dose is further increased to be 8-10 times of the dose, the treatment effect is inferior to that of single-use treatment effect of the two medicines. On the basis of the optional best dose, the total effect of ginkgo biloba leaf extract and donepezil in different weight proportions is further evaluated, and the weight of the two medicines is found to begreater in proportion of ginkgo biloba leaf extract or donepezil, so that the total effect is in downtrend. According to the research results, the Alzheimer's disease prevention effect is optimal when the weight proportion of the inkgo biloba extract to donepezil is (1 to 1)-(1 to 3), so that very good synergistic interaction is achieved.
