[0039] 1 Materials and methods

[0040] 1.1 Chemical reagents and instruments

[0041] n-Hexane was purchased from Shandong Hongyan Reagent Factory. DAS, CAS 592-88-1, ≥97%, purchased from Sigma-Aldrich Company. Corn oil was purchased from Ginza Supermarket. The rotary rod tester ZS-ROM was purchased from Beijing Zhongshi Dichuang Company.

[0042] 1.2 Animal handling

[0043]50 adult male Wistar rats were purchased from the Animal Experiment Center of Shandong University, weighing 180-220g, and randomly divided into 5 groups: blank control group, DAS control group (100mg / kg.bw), n-hexane model group, DAS low-dose intervention group (50mg / kg.bw), DAS high-dose intervention group (100mg / kg.bw). The room temperature was maintained at 20±2°C, the humidity was maintained at 50±10%, the day and night were alternated for 12 hours, and the supply of water and food was maintained. After one week of adaptive feeding, the DAS was prepared into the corresponding concentration with ...

Abstract

The invention provides the application of diallyl monosulfide in preventing / or treating peripheral nerve injury caused by n-hexane, belonging to the technical field of pharmaceutical preparations. Theinvention adopts diallyl sulfide to interfere with peripheral neuropathy induced by rats exposed to n-hexane, and evaluates the intervention effect. The results showed that diallyl sulfide had a goodeffect on peripheral neuropathy induced by n-hexane poisoning in rats. Moreover, the FDA proposed in 2005 that diallyl sulfide can be used as a food additive, so it is very suitable for the long-termuse of n-hexane occupational exposure population. The research of the present invention is helpful for exploring the mechanism of the above results, and has important clinical application value in preventing and treating peripheral nerve damage caused by n-hexane; and the invention opens up a new pharmaceutical use for diallyl monosulfide and other clinically safe bioactive ingredient of garlic.

Description

technical field [0001] The invention belongs to the technical field of pharmaceutical preparations, and in particular relates to the application of diallyl monosulfide in preventing / or treating peripheral nerve damage caused by n-hexane. Background technique [0002] n-Hexane is a commonly used industrial solvent and itself is a low toxicity substance. However, after n-hexane enters the body, it will form the final poison 2,5-hexanedione under the catalysis of cytochrome P450 in the liver, and then induce a series of peripheral neuropathy. Its main neurotoxicity is to cause swelling of the shaft near the distal end of the peripheral nerve, and its pathological features are axonal neurofilament accumulation and segmental swelling, followed by segmental demyelination, with long and thick fibers being the most obvious. Many scholars have studied the pathogenesis of n-hexane-toxic peripheral neuropathy from the aspects of axonal skeleton proteins, energy metabolism disorders, ...
