Problems solved by technology

The cost of silicon chips is high in the manufacturing process, and a large amount of harmful chemicals will be used, such as silicon tetrachloride, cyanide, dichloroethane, etc., and a large amount of water resources and fossil fuels will be consumed. According to statistics, about one-third 1 in 2 workers suffer from diseases caused by silicon pollution
Secondly, the larger the capacity of storing information, the larger the volume of the storage device, and there will be a lot of inconvenience in carrying, storing and maintaining the device. Moreover, discarded devices are not easy to degrade and easily cause environmental pollution. Silicon is also a non-renewable resource. a day will be exhausted

Embodiment

[0029] 1. Sequence Synthesis

[0030] The three poems of Cangyang Gyatso are coded into DNA sequences, and then the DNA is artificially synthesized, PCR amplified with specific primers, sequenced by Sanger method, and finally the text is decoded and restored. Taking the whole process as an example to explain in detail a method using DNA as A method for efficiently storing text information on a medium.

[0031] All Chinese characters and punctuation marks in the three poems of Tsangyang Gyatso have a total of 115 characters, a size of 11kb, and a total of 11364 bits. The DNA information sequence will add primer binding regions at both ends and sequence protection regions at both ends. The final artificially synthesized DNA fragment comprises a primer-binding segment, a sequence information protection segment, and an information segment ( figure 1 ). The content of the three poems by Tsangyang Gyatso is short, so it is set as a storage unit to synthesize a DNA sequence with a...

Abstract

The invention provides a method for taking DNA as an efficient text information storage medium, which is used for storing text information and is beneficial to culture protection. The method for taking DNA as a text information efficient storage medium comprises a storage process and a decoding process. The storage process comprises the following steps: establishing an information sequence, dividing text content into a plurality of storage units, and converting each storage unit into a DNA sequence by utilizing DNA tool software, thereby converting the whole text into a plurality of DNA sequences; adding 20bp protection sequences to the two ends of the information sequence respectively; adding primer binding regions at two ends of the protection sequence. The decoding process comprises thefollowing steps: culturing bacteria, extracting plasmids, carrying out polymerase chain reaction to obtain an amplification product, carrying out agarose gel electrophoresis and imaging identification on the amplification product, sequencing the amplification product by using a Sanger sequencing method, and reducing the DNA sequence obtained by sequencing into a Chinese character text by using DNATools software.

Description

technical field [0001] The invention relates to the technical field of DNA storage, in particular to a method for using DNA as an efficient storage medium for text information. Background technique [0002] The 21st century is an era of rapid development of information technology, and the amount of information is increasing exponentially. The long-term stable storage of this information has become the focus of attention. The current devices used for data storage such as paper, magnetic media and silicon chips are not ideal storage devices. First of all, the data storage density of silicon chips is limited, and the storage quality of tapes will decline rapidly as the number of times of use increases. The cost of silicon chips is high in the manufacturing process, and a large amount of harmful chemicals will be used, such as silicon tetrachloride, cyanide, dichloroethane, etc., and a large amount of water resources and fossil fuels will be consumed. According to statistics, ab...
